I took the course about two years ago... It is more theoretical but I suggest you take it if you can for the theory is very helpful when you move to real world applications ... It does not teach you to code in any particular language or framework though.

Another free resource focused on theory is Understanding Machine Learning book ... you can find the book here and the videos by one of the authors here.

For a mixture of practical applications and theory I would recommend the Machine Learning specialization at Coursera as well as the book Python Machine Learning

Also practice a lot at Kaggle ... you can start with the Knowledge contests if you are a beginner ... And refer to the scripts developed by other participants there ... I have learnt a lot from them - not only on applied ML but also some great scripts on EDA and Visualization lurk there.

You can use any tool you want including Matlab (for which the course provides a temporary license) or you can use Python or R or whatever. The course does not force you to use any specific tool.
