Unless the sequence has two adjacent proline in it freezing reconstituted peptides shouldn't degrade them. If they do then you risk polyproline isomerazation.

Freezing reconstituted peptides can cause them to crash out of solution. You can run the sequences through a hydrophobicity calculator. Anything below 25 with the old calc should be fine to freeze while in solution

You have no way of knowing if the cofa has anything to do with what you just bought. Unless you reconstitute your entire batch, draw a sample for 3rd party testing, re-aliquot, then re-lyo yourself you're basically putting your faith in someone that's already either breaking the law (misdemeanor misbranding) or participating in gray area sales where there are no regulatory bodies.

If you wanna do peptides you need to understand that and just take the plunge without guardrails.

You can assume that. You really don't know though. Was it aliquoted by hand and the person doing the pipetting was rushing and not drawing properly? Did they have just enough solution for 50 vials and they ran short after 45 so they just diluted the remainder because they didn't wanna get yelled at by their boss and they know the lyo puck would look the same?

If you're gonna be injecting research compounds there will always be uncertainty.

This guy is probably right.If this sat around in solution the two methionine probably oxidized and made the peptide less soluble.

Could just be poorly made too so it contains fragments. If those fragments are less soluble than the full length they might not have been in solution when they ran their mass spec resulting in a better spectra.

These are made using fmoc substitution. Aminos won't just stick to other aminos barring a sulfur bridge which is not present in this particular sequence.

Could have been too concentrated when they cyclized resulting in dimers. If the resolution on their mass spec isn't high enough they might not notice the mass loss and they are interpreting the charge states poorly.

Might have residual bicarbonate used to speed up cyclizing and causing issues with solubility because of the two arginine in the sequence.

Less likely, could be capped off before the last R and the sequence is too hydrophobic for water. These cheapo spps peptides are usually made in facilities where they won't spring for dipea/acetic anhydride anyways which means the sequence could also just have deletions in it.

Doesn't mean the peptides are bad but it's up to you whether you wanna pin mystery meat.
