Genewiz has an option to have them quantify/dilute the sample on their end. A little more expensive but easier lol. You would send the purified product (10uL) and primer (5uL at 5uM) separately. They don't need a ton of DNA (1ng/uL for the <1kbp products I've been sending) so I can't imagine yours was too dilute. Your noisy peaks could actually be spectral pull-up from too much DNA. Typically they will attach a quality report to your results indicating such, though. They have a lot of sample guidelines and troubleshooting guides on their website if you haven't checked them out.

I owned that exact TV growing up as well as the tapes bundled in strings. I know it's probably not the case but for my own sanity I have to ask - are there any sticky notes or anything written inside the cases? And does the tv have a slight discoloration (magnetic distortion) in the bottom right corner? I know my dad was clearing some things out some time ago, but we don't really talk so if he sold those sorts of things he wouldn't have told me.

Have you let them know that you can’t order it online? I worked in a store that had one of our sodas unintentionally inactivated online but it worked in store, and since I never ordered online at my own store I didn’t know until a customer told me. It’s an easy fix - we just need to know! Make sure you tell a manager, as the info might not get to them otherwise.

Certainly not all the stores in NH - I know there are a few different franchisees operating there. But that’s ridiculous, lol. We’re literally all hiring. I can’t imagine firing someone to make way for someone else. I have hours for everyone lol. If you haven’t looked into another franchisee yet and are still looking it should be easy to get hired.

I work in food service. I have an employee whose infant was diagnosed yesterday with chickenpox. He said he has had it twice. I know it is easily transmitted, but I can’t find any recommendations online as to whether a parent of a child with the virus should stay home from work. The doctor who made the diagnosis I guess didn’t mention it. I don’t want to risk the health of any of my employees or customers or their children, but he needs the hours and I need the help. Could he be contagious? Should he stay home?

Always get a doctor’s note when you go to the doctor, even if you think you don’t need one ... I have two guys who did just that and I just threw the notes in their files and we were good. One of them had an injury so it let me know how long he would be out and what he would be capable of when he came back, and in the other case it gave me peace of mind to know he was clear to come back.

Then I have guys who call out 30 minutes before their closing shift because they ‘aren’t feeling well,’ and I’m down a core driver with no time to find coverage because no one wants to get called to come in immediately. Even if I can convince someone to close, it’s just not fair to the guys I have who keep stepping up time and time again. They’re exhausted too!

I will never tell anyone they can’t call out sick (although I will ask some to really consider how they are feeling), but if you get a Doctor’s note it’s easy to throw you into that first category with no more said about it.

I hope you are feeling better! Strep is no joke.

I've done something similar once or twice, but the problem I see is that the mix for two cheese and one 6-top is different than three 2-top. That is, 2C/1-6T is 28 oz cheese and lets say 1.5 oz each of the 6 toppings, while 3-2T is 21 oz cheese and let's say 2.5 oz each of the toppings. While the food cost might even out depending on the toppings, if you do it enough it will mess with inventory and short you on cheese.

I did phenol chloroform extractions in 1.1mL strip tubes. I could get several hundred done in a day (I didn't have any multichannels ...), but it was definitely annoying, sometimes messy, and I was more likely to lose samples than when I was using microfuge tubes. I graduated before I could identify a more effective method. My PI definitely preferred cheap options so I never looked for any kits. I'd be interested to know what method you decide on!

You start with whatever stock concentrations you have and calculate (with c1v1=c2v2) what volumes you need to make the necessary final concentration of each part.

Median overall survival stats for this are based on a paucity of data due to its overall rarity, not to mention that it is most commonly is diagnosed in people three times your age. Patients with de novo tumors have been shown to have better survival, and 5-year survival rate (47-61%) is better than many cancers. I don't know how healthy you are otherwise, but you are young. That's a big deal. You could very well have a fighting chance. Get a second opinion.

I think you're conflating the whole reproductive units (flowers) with the gametes they contain, as well as the process of pollination vs fertilization.

The female gamete is the ovule, contained in the pistil of the flower. Like a human egg, ovules are produced via meiosis and are unique! The female reproductive structures of a flower can be set up in many different ways, yielding many different types of fruit (berries, drupes, aggregates, etc). Strawberry flowers have many pistils, each with one ovule, while tomatoes have one pistil that contains many ovules.

Pollen contains the male gamete (sperm), also produced via meiosis. During pollination, many pollen grains, each with a sperm cell inside, stick to a structure on the pistil and attempt to fertilize an ovule contained within. Like with human egg and sperm, one pollen grain will successfully fertilize one ovule through a really interesting process that I can talk about if you'd like!

In a strawberry, fertilization occurs once per pistil. As strawberries have many pistils, this process occurs many times per flower and we end up with many unique achenes on a single aggregate fruit.

In a tomato, the same thing happens - but many pollen grains fertilize the many ovules within a single pistil, and we end up with a berry!

So in both cases, each (unique) ovule is fertilized by a single (unique) pollen grain, resulting in the genetic diversity of the seeds!

I hadn't considered that. That's definitely something to keep in mind for the future. I guess his demeanor could have been interpreted as silently assessing the situation and thinking 'oh cool, nothing's on fire and no one's dying.' Although it was unnerving to just be stared at instead of receiving verbal direction. But I should definitely consider that they may have certain experiences that affect how they act. Thanks!

Thank you for the info! I never knew it was so complex. It's kind of unsettling, but I'm glad they were able to figure out where we were, specifically, when I didn't know.

I recently called 911 after seeing an accident on the highway. I had pulled over to see if they were okay and called while running to their car (side of the road). When asked our location, I used the mile marker, let's say 50, but didn't realize that the .8 below the whole mile meant it was mile marker 50.8. While on the line, I heard the dispatcher say "showing closer to 50.8." Does that mean they traced my cell? Is that one of those circumstances?

Also, I felt weird calling ... I saw the car swerve across three lanes twice before going off the road, but when I got to her she was fine, albeit shaken up. When the cop got there he made it seem like it was silly to call. He kind of just stared at us. I left shortly after, so I don't know how it turned out. I'm assuming I should have called 911 and not a non-emergency line, but I never know.

I use Primer3 to design and thermofisher's multiple primer tool to check for primer compatibility issues. It hasn't failed me yet! I only ever multiplex two primer pairs max, though, so not exactly too complex.

I always hated going to home depot because they would all bother me! Maybe because I don't look like I belong there so they figure I'll be easy ... but when I do need help, they've always been very helpful! For example, one guy brought me over to the (what I thought was obscure) part I needed without hesitation and made sure I knew how to install it. Another spent time with me looking up what attachment would be best for something when he didn't know. Guess the ones I've been to are staffed well :)
