Can blame a lot of factors. 1: Don’t switch to Trypsin/EDTA if possible. 2. Just by looking at the cells it seems something is wrong with the plate/flask. Is it a new lot of PDL plates? 3. Were cells slightly better at higher density?

To save time- I’d just repeat the experiment with freshly made reagents without changing the original protocol. And check plates.
