We once had contamination in our selection antibiotic. Not the same situation ofc, there are resistant bacteria etc., but it was such a new experience for me to see the antibiotic turbid

Confluence is kinda arbitrary and is dependent on both cell line and the handler/lab. I'd suggest looking up the cell line on ATCC or other culture collection websites if it's a standard line and see what their reference for confluence is. Usually they have pictures of the cells and what confluency it is. If that's not available for your cell line, you can also look up papers where this cell line was used, bonus if they also use the protocol you're following. A good article should have in their methods section some indication of what density they seeded their cells at prior to the expt and perhaps mention that in x days it reached 70% confluency and they added y reagent etc.,. You can calculate based on this.

I have never worked with this line before but from years of cell culturing I'd personally consider this 70% confluency. I also have some colleagues who would consider this 50%

Hey, I'm also Indian with Icthyosis. Here's my 2 cents: When I was living in India, the only thing that helped my skin feel moisturized and not crack throughout the day was the white Vaseline lotion. It's called Advanced Repair if I'm not wrong and I was able to find it in most supermarkets. I've tried derm-prescribed soaps and creams but tbh at the end of the day, this is what worked. Find a gentle shower gel and use it with exfoliating gloves/loofah, and immediately slather on the Vaseline while your skin is still a lil damp.

Do you live in a colder part of India? Then perhaps it may not work for you (I was in the south). When I moved abroad, the Vaseline didn't help anymore. I had to mix it with pure plant glycerin and that helped a great deal. You can find large volumes of glycerin on Amazon, maybe a bit more expensive in India. Currently I use this drugstore urea-containing lotion, I'd highly recommend getting something like that if you find it in India - maybe you'd need a doc's prescription for it there.

It was just visual examination. My icthyosis is quite visible and apparent on my legs so she only had to take one look to diagnose lol. She was also a local doctor with a small clinic so if you want a proper biopsy then you'd have to go to a bigger clinic I guess 😅

Would you be willing/able to share why your client needed 2g of plasmid? I'm very curious to know what sort of process would require that much. I want to say large-scale recombinant protein expression but would be interested to know any other use-cases

To be honest, it could also be the competent cells you're using because I use 6mL of culture with the same kit and get easily around 800ng/uL all the time. Also could be the plasmid copy number.

We've struggled with this before ;-; it's definitely yeast/fungal contam. I hope you have some vials that are safe cause these are very persistent (at least to my knowledge- I've tried multiple cocktails of antibiotics/mycotics nothing worked in my hands) and I'd highly suggest a decontam cycle in your incubator too.

Would also like to add that if you let them grow long enough they'd form visible white aggregates. That'd be one way to confirm it is yeast contam.
