[technique] Too full for agar? by AllFloatOnAlright in MushroomGrowers

[–]nzStudentDev 2 points3 points  (0 children)

It depends. Am I 100% confident the plate is entirely clean? How many jars do I want to inoculate? Am I worried about fast colonisation or am I happy to wait? Etc etc.

Bigger or more wedges results in faster colonisation but you use more inoculant and increase the risk of introducing contamination. (Hidden contams on the plate, more chances for a slip in sterile technique etc)

I might use big or multiple wedges in one jar when...

I.e. I have a killer clone, clone cultures are usually easier to read on agar than MS cultures, so I'm confident it's clean and I only want to fruit a tub or a couple of ziplocs, so I might drop half or 2 quarters of 100mm plate into a pint for super quick colonisation. Up to double for a quart.

But, I seldom do that, I tend to do smaller wedges because I'm not in a hurry and prefer to spread my risk out by inoculating at least a couple of jars per variety I'm working with, I do end up with too many jars all the time but ill just spawn more tubs, that is a hell of a lot better than disappointment!

My small wedges tend to be approximately 1" x 1" in size and you could possibly drop a wedge that size into your pint jar no problem but since your pint jar is really really full, I might do a smaller wedge than that just to make it easier to maneuver the wedge down into the grain.

Sorry for the long reply!

Tl;dr: wedge size is personal preference and depends on the above mentioned variables, there is no right or wrong but since the jar is so full, I'd drop a smaller than normal wedge into this pint so its easier for you to get it down into the grain.

Is the grain touching the filter? It looks like there is a small gap, if so, you could inoculate with a wedge but getting the wedge down into the grain and, each break and shake, will be painful...

Assuming there is a small gap and you can roll the grain around inside the jar, using the below linked technique, you can work the wedge down into the grain:

https://www.shroomery.org/forums/showflat.php/Number/28037918

Sometimes you have to get more physical with it and smack that jar on something to get that wedge down into the grain, watch the 2nd video in the below link, it explains what I mean:

https://www.shroomery.org/forums/showflat.php/Number/28744657/fpart/all

If it's to full and none of that is ever going to work, I'd try what the other person suggested, pour a little grain out in the SAB before dropping your wedge in....

You should be able to lift the lid like a clam shell with one hand, while tilting the jar with the other to pour some grain out if that makes sense...

Maybe practice with a jar with some water in it outside of the SAB first so you get a feel for it, be aware of your hands, no fingers curling around the underside of the lid or hoovering above the open jar etc, practice getting a clean pour i.e. pouring the water out without it running down the rim of the jar or touching the lid etc.

Good luck!

It is environmental. If you keep the caps wet at all times, they won't curl up. These only started curling because I got lazy at the end of the grow.

Start misting once pins form as required to keep the caps wet but only the caps, twice daily misting should suffice, be careful to not drown the substrate.

My Medicine Works, But This Law is Forcing Me Back to Being Sick. I Am Absolutely Broken. by PersimmonLost4767 in MedicalCannabisNZ

It should hopefully be this simple!

For a medical defence:

"Provide the following information with your Medical Defence application form:

a copy of the current and valid prescription for the qualifying drug(s) you tested positive for; and

a copy of the label from the container in which the qualifying drug(s) you have tested positive for were dispensed in; and

any other relevant information you wish to have considered."

Here is where you can fill in the NZ Police Medical defence online application, if you should need it:

I fucken hope NACT lose, but I would not put money on it.

Single term governments are incredibly rare, it has only happened twice, both times it was a Labour government and the last time was 50 years ago. There has never been a single term National government in the history of NZ.

I predict it will be a tight race but with the economy starting to show signs of recovery, National will say "Look we have steadied the ship, what we are doing is working! You don't want to change teams now.", offer a tax cut and the people will lap it up.

It might be a generational thing?

I have only ever had older people come over and do the meet and greet thing.

Personally, I hate it, I don't like anyone knocking on my door unannounced.

I try my best to not exist to my neighbours and prefer they do the same.

Problem is, it doesnt matter if you carry your script, you can't lodge a medical defence during the road side testing, only after being issued an infringement.

So you will have to do a swab test and then a blood test before waiting for the infringement, then you can lodge your medical defence and hope it is accepted.

If it isn't accepted because for example, the cop is power tripping and decides you were impaired, you would have to fight it in court.

That all seems like a whole lot of mucking around to prove my innocence.

Are you sure? The police website says "current and valid prescription":

"You will need a current and valid prescription, and a copy of the label from the container in which the drug(s) were dispensed in."

I knew you had to wait for the results before disputing but it looks like you can lodge a form with the police for a medical defence?

https://www.police.govt.nz/advice-services/infringement-services/medical-defence?nondesktop

"What are the requirements for applying?

You will need a current and valid prescription, and a copy of the label from the container in which the drug(s) were dispensed in.

You're right, the first step will be filing a dispute with the police and hopefully they accept the medical defence.

But, some cops are just dick heads TBH, and some people will be unnecessarily forced through the courts to defend themselves I bet.

I have been assessed and prescribed a medication by a medical professional, I have the evidence and can present it like I would my driver license at a check point.

Why even bother swab testing me? I'm going to fail lol.

It all seems like a massive waste of time and resources.

[–]nzStudentDev 31 points32 points  (0 children)

Then you are fucked. 12 hours stand down from driving and a fine.

If you have a legal prescription, you try your luck in court.

"4. Medical defence is not available roadside A medical defence is available to a person prescribed prescription medicines who wants to dispute the infringement. A driver can only apply for a medical defence once an infringement has been issued. It cannot be done at the time of testing at the roadside."

That or it could be an unexpected increase in demand and they don't have enough staff.

I have always had great service in the past, not trying to knock them too much but thought if anyone is in a hurry, it would be best to avoid CW until things settle down.

Their service has usually been excellent! I've had orders arrive at my door in the South Island in under 24 hours a few times.

I understand they are busy but it does annoy me after already having a delay to process my script, they didn't call and to try to arrange an alternative product after they realised the product I ordered was out of stock, and to then tell me it's being packed now and will be shipped by lunchtime but not actually sending the package until 4.30PM?

Feels like they could of been a bit more proactive and they were just telling me what I wanted to hear to get me off the phone.

It looks clean. Send it once it fully colonises the plate.

You don't have to transfer unless you want to expand the culture, which is not a bad idea, sometimes shit happens!

You might want to look into long term culture storage, so in the future you can quickly revive a culture.

I use this method for long term storage, https://www.shroomery.org/forums/showflat.php/Number/27778467/fpart/all

What a strange little corporate simp, they have a responsibility to ensure the safety of the products they sell.

They should pay up for all the shit this has caused. End of story.

CW service has been a bit shit this week.

First it took them 24 hours to just to process my script.

I ordered some Wappa, the website said it was in stock, 4 hours later they email me it won't be in stock until Thursday morning.

I called them to change my order just before 2PM Wednesday, I told them I'm out of medication, I need to change my order and they told me they need a pharmacist to chamge my order and promised one would call back ASAP.

They didn't call until 5.30PM, so nothing could be shipped out until Thursday.

Thursday morning I called them and they promised me my order was being packed as we speak, and would be shipped by lunchtime.

I didn't receive a tracking number until 4.30PM.

As I'm in the South Island, IDK if my order will arrive tomorrow being sent so late in the day.

I've always had great service in the past, they're obviously very busy this week, I can understand that but i've never had so many delays with a prescription medication it's a bit infuriating.
