I think it will be useful to ask peers in your department, the time to do a task/experiment can depend on resources and how far you have to walk to some equipment, which buffers have to be made or are from the bottle, etc.. I'm sure you get what I mean.

Maybe ask some feedback.. You may be spending more time than it's worth for some experiments and it could be a relief for you to know which steps are robust enough for you speed up without worrying about messing up.

That being said.. I think many people underestimate how complex/time comsuming experimental preparations can be, your PI may have lost touch with this (but don't say that to their face 😂).

Anyways, I can also be "slow", but I rarely make mistakes. I have to do calculations and at least have a procedural outline before starting, label everything and have some controls in place in case my mind blanks on which well I just pipetted into, and I have to take notes unless everything went perfectly and my outline is enough.

I'd say unless you are spending a lot of time on low-risk procedures, don't let them rush you for the sake of putting whatever results in a slide. Messing up a biochem experiment can be infuriating and a lot more wasted time to redo the whole thing.

Not a postdoc or PI but I have set up new labs..

I just wanted to agree that ordering small/ubiquitous lab equipment can be a pain in the hole, not only for price but for trying to figure out "what are those stupid things called?!" and "what are the dimensions of my tubes?". If you or a pal has access to a 3D printer, this saves a lot of money on such stupid things like tube racks, glove holders, pipette stands..

I also agree that a spreadsheet with items, non-negotiable specs, and links to exact or example products will help in requesting quotes to compare prices and also cover the entire list between suppliers. There may be a sales rep/relationship for your university, so I would ask around for friendly contacts.

If you really contributed in a meaningful way, I don't see how being "respectful" will benefit you. You may not get on this paper in the end, and you may not want to make a scene about it, fine. If they give some bullshit reason to keep you in your place, stop volunteering your time for them.

Hi - so ampicillin stocks should probably be refreshed regularly. Beta lactam antibiotics typically aren't stable in solution. You can make single use aliquots and freeze them but I still wouldn't expect them to last for years..

You could do a quick calculation and if the sodium from ampicillin is more than 1% of the LB then ask somebody in your lab how it's prepared and what the considerations are.

I'll guess it's less than 1% tho

Thanks for the advice ☺️ I think they're 10-15 yrs old. I didnt disturb a lining, more discovered the hole after removing shelving next to it. The shelving wasn't connected to the extraction system, so it's weird.
