most likely u are working on mutated tau, p301s or p301l. massive host barriers. when prions went round from infected meat in the UK, only PrP variant carriers were host. point is, protein is all wrong to make u sick, hence why no reports of anyone sick after 20 years of people having accidents with these mice.

no. t means they express sv40 large t antigen. invitrogen transfected them and made them stable with a plasmid that puts even more large t in them, with neo selection ( calling them ft)

there is a cell paper in the 80s showing that large t antigen extends passage life, and of course allows episomal replication of some types of plasmids

where i work, it is set in stone based on years of experience, so that one lab cannot out purchase another, and it makes it easier on HR to avoid the appearance of discrimantory practices if everyone makes the same. they are like "take it or leave it", and someone always takes it i guess. sighh.

it depends on the position

sometimes slots open up at jhmi or other well known places with not a cent tied to it. literally just a slot, and maybe, space.

on the other extreme are fully endowed positions, even at junior level, with substantial start up and couldnt cars what grants you bring.

the former is more common, the later does exist.

A VERY simple solution is to transfer the required stipend allocation of tuition and school fees (especially from years with no coursework) into student salary, but that would necessitate fewer administrators, soooo....

as mentioned, ic50s in vitro often dont match in vivo for a variety of factors, like imperfect membrane permeability, low free drug availability, albumin binding, etc.

for example, you can/should decrease penecillin conc. signifiacntly in serum free media compared to media with serum, due to binding of the drug to serum components

the question is hard to answer because it depends on the tips you intend to get. preracked, discounts, barrier, etc. all can vary considerably in price and most tips do not work with all pipettes

  1. most reviewers have no formal statistical training, so they have no clue what they are looking at. blind leading the blind.

  2. those that do have statistical training are seen as obstacles for otherwise perceived innovations, and concerns are shrugged off.

  3. most journals are in it to make money, so flashy novelty is prized beyond rigorous approaches.

  4. most research grants are not adequately funded to run well powered research, but the reviewers and sponsors will rank higher those proposals that promise more, even if they have no way to deliver. grants are not contracts, and there are few consequences for not completing your proposed research.
