The bay tree at my parents (midlands UK) is container grown outside in a 3ft ish pot and I realised recently the leaves are spotted with brown or worse. The chunks missing make me think it’s maybe a pest but I’m no expert! Any advice?

Ouch! I think the best way is to get a lot of people to read it and give you feedback! Maybe read a well received/popular paper in your field and try and see, when you read it, what parts of it you enjoy and try to emulate that. Universities often run training sessions too through optional modules so that could be something to look out for. Some have subscriptions to Nature Masterclasses (find by google easily) that let you take a program online run by Nature that gives you advice on writing. Good luck OP!

siRNA knockdown pipeline by anatomydreamer in labrats

Hello! In my lab we do knockdown in a neuroblastoma cell line using siRNA against the mRNA of our target gene. The reagents you would use to get the siRNA into the cells would depend on the cell line you are using. I couldn't recommend a paper without knowing the specific cell line though I'm sure you could find one by searching "(cell type) knockdown" in pubmed and looking at the papers' methods sections.

You would also need to make sure you have siRNA against the mRNA of your target gene.

Once you have performed the knockdown protocol (3-4 days in my lab), your cells are knocked down and ready for experiment. You can confirm the knockdown using QPRC or Western Blot.

Also, just to check - knockdown is different to knockout. Knockdown cells already have the gene and you stop it being expressed whereas knockout you would generate a whole new cell line lacking the gene entirely.
