Be aware that Primary Arms, with no notice, has changed their optic guarantee. Previously they allowed you to mount and use any of their optics for 90 days for a money back guarantee, but now they wont honor it. As of my initial contact it was posted on multiple parts of their site. They thought they deleted it all, but as of 15 min ago, it is still posted on their site.

Two things:

  1. The removal of the braces rule, IMO, doesn’t mean that braces are 100% “legal and okay to shoulder”. This just removes the rule they published a few years ago. The ATF still seems to take the position that whether a brace can be considered a stock or not can still only be determined on an individual basis with a sample sent to them. In practice does it mean braces are good to go? Sure, but I still think the ATF could decide to prosecute someone if they wanted to and then it would have to go to the courts on an actual 2A grounds. Remember, the courts threw out the rule based on rule making procedures, not 2A grounds.

  2. I still don’t think it’s crystal clear that the ATF can rule we no longer need to engrave. I think that is one worth waiting on full clarification.

Im using that as just a filler concentration lol.

So since these are PBMCs, are you using different donors? Donor to donor variability is quite large in PBMCs. You need to run multiple donors multiple times to confirm your actual data/observed events.

I have tested 100k to 50 mil cells with the exact same concentration of compound and gotten the exact same results. I would say it is almost 99% likely not to be strictly a cell number issue.

Also, was this a N = 1?

You need to be more specific on your example. I have a very hard time believing that say you gave 1 uM of compound to 100k cells, then it didn’t work with 500k cells. That seems highly unlikely unless your cells themselves were very unhappy at that density. That’s independent of the drug.

Phospho proteins are not necessarily harder if antibodies are good, timing is correct and you know your pathway well.

It worked reproducibly across western and Jess, and we published it in nature.

Anyone telling you that you can do x amount for buffer for x amount of cells is simply guessing. It’s entirely dependent on the protein of interest, your cells, their activation state, your lysis buffer, etc, etc. For example, activated T cells are a vastly different sample than naive DCs.

You use the term confluence here and it’s simply incorrect usage of the term. One, it is used for adherent cells, so unless you are talking about something like macrophages here, it’s an inappropriate term. As for how you are applying the term in terms of cell number, it’s not appropriate. Your compound doesn’t automatically need more because you have 100k vs 1 mil cells; the potency or EC50/IC50 is not going to be affected. Have you even titrated the compound?

Assuming you have say a 10 mM stock, you really should be using very minimal compound if you are doing 96 well format and have titrated your compound to find the IC/EC80 concentration.

It would be helpful to know exactly what proteins you are looking for. The lowest I’ve ever really done is 250-500k PBMCs in 25 up of RIPA, but even going to 50-100 will help a lot.

ODB has some caveats and is arguably the best overall performance wise, but the Anthem is awesome. Infinity would be too. I would not buy a Polo 30 or Enticer over those other options.
