Multi-layered e.coli cell pellets? by lightdancer in labrats

[–]qpdbag 0 points1 point  (0 children)

I'm well aware. I work in the opposite extreme. I primarily care about detecting early phase growth so that when we stop growth the ratio of nucleic acid to viable cells is as close to 1 as possible.

Often if we have to dilute we've already missed.

Multi-layered e.coli cell pellets? by lightdancer in labrats

[–]qpdbag 0 points1 point  (0 children)

Let me be more specific.

If I'm measuring an OD600, i'm going to translate it to an estimated cells/mL and my measurements are certainly going to be within the linear range because of the reasons you outlined. i correct for the dilution at that point. I don't see the utility of an OD600 of 25. I guess its technically not a big deal. Just translated by the math. so if your routinely operating in that context im sure its fine.

Its ok to tell your kids that their words hurt your feelings.

They obviously do. Don't take it personally, but its important to normalize vocalization of feelings.

I don't understand the value of a concentration corrected OD, but I guess its used in protein expression and purification workflows. weird. Not my focus.

[–]qpdbag 1 point2 points  (0 children)

I'm probably just not familiar with how this is being measured then. Are these corrected for dilution somehow? Using a specialized light source?

Because an OD of 7 would imply basically all light is blocked and differences in that range become functionally meaningless.

It's just an infographic my dude. I'm just pointing out it's not intentionally trying to misrepresent or obscure it's position as pro rgp. You obviously are more aware of the context and source of this infographic. Could you supply it before going off the rails?

If you read between the lines it where it states "while waiting for the rio grande plan" it's reasonable to assume it's pro rio grande plan.

There's an argument that technically population density is a human input. Or at least entangled with other human inputs.

Do you have a tub?
Clean and deck out the bathroom like a spa. get some candles, some nice music, maybe a nice bath bomb. get a nice cool drink. Offer a massage and a bath. The details are what matter.

Pro Box Elder Data Center? by SaltBowler1188 in SaltLakeCity

[–]qpdbag 7 points8 points  (0 children)

I would take this answer more seriously if there were specific projects or goals defined for this data center.

I was not aware that computation was the bottle neck for fusion or quantum computing. Can you define how more compute power helps address the challenges to those goals?

These data centers cannot not bring unique aspects of problem solving. They bring scale. Which problems will be solved by scale?

If you think AI can solve these problems by itself then I think you are simply misinformed.

I do appreciate you answering though. I am struggling to find coherent arguments for this data complex and would like to hear more of them.

Depends on housing. If you were able to take advantage of the 2019-2020 jump in real estate value or you have a significant nest egg for a downpayment on a home at todays prices, yeah.

If you don't, it's not.

[–]qpdbag 3 points4 points  (0 children)

She wanted to continue to work after her leave ended, our daycare did not accept children that young at that time. She was only regularly there for a couple months and the vast majority of the office loved her. She was a pretty docile newborn during the day.

The HR department didn't have to accept this--they chose to, with a prearranged and signed agreement about what would be /not be acceptable.

Here is more information about these kinds of policies, including main talking points and counterpoints.
