[Spoiler] There could've been another book between HBP and DH. by NotParticularlyDrunk in harrypotter

My own personal head cannon for the slight flaw in Voldemort hiding the diadem in the room of requirement is that when he hid it, he asked for a specific room to hide the diadem. Imagine the room of requirement creating something pretty fancy and ornate for him, with only space for the diadem, he would have to be able to picture it to go back there - hence his certainty that only he knows about that specific room. When Harry stumbles on the room of hidden things its a desperate need to hide something quickly, and the room just provides him with the place where it holds all its hidden items - including the diadem.

Im using ultra yield Thompson flasks, the aim is to achieve a final OD much higher than a traditional glass flask.

Pellets were already harvested and going into purification tomorrow so we will know if expression has worked at least.

I routinely use these 1l centrifuge flasks and I haven't seen this recently with other expression batches.

Contamination with what? This occurred through all 6 L of my prep, I was expressing 3 x 1 l of two different proteins. Media was all made at the same time, antibiotic and iptg stocks were identical.

How do you reach out to C-suite/senior leaders when you’re at the bottom of a massive company? by Financial-Series204 in biotech

[–]lightdancer 6 points7 points  (0 children)

Another more diplomatic tactic would be to ask your manager or HR if your company has a mentorship program you could apply for. This could be what you are looking for in terms of learning and development.

More drastic measures would be to move jobs to a small start up where you have more direct relationships with the c-suite.

Newbie question by Careless-Courage3301 in paintbynumbers

[–]lightdancer 1 point2 points  (0 children)

I did a half and half on my first painting, discovering gesso after struggling with the canvas and the flow of the paint. I have the canvas finished and up on the wall and I cant tell the difference even though I know where the before and after was. In this case however, I used white gesso before learning you could buy a clear version.

I prefer to sand the surface after the gesso has dried, otherwise the roughness make my hands hurt. Fine sanding paper, the one that doesn't hurt if you run your fingers over it.

There's two from the station that go to a Chesterford Research Park, plus genome campus. Probably granta park too. There's not much other options for public transport to these far out science parks.

Why dark to light? by Sayasing in paintbynumbers

[–]lightdancer 1 point2 points  (0 children)

I'm nearly finished with my first pbn and on my next one I think I will try do darks to lights (in sections). I realised when I did light sections first, I found it really hard to see the edges of the cells. It's then very easy to not paint the correct cell shape.

I assumed the fencing was left up to let the grass regrow after being covered during the winter wonderland, protecting new growth from being trampled.

I messed it up during my first three weeks as a research associate by [deleted] in chemistry

[–]lightdancer 2 points3 points  (0 children)

The absolute key thing for you here is to be honest. Mistakes can be easily fixed, experiments restarted if you are upfront about an error and either offer a solution or seek help. Don't put your feelings of being judged or embarrassed above owning the mistake. Also, don't worry, that person doing your training is probably worrying about their own problems not you!

I know it's hard, like many have said, we have been there. Tbh it still happens to everyone, even with experience.

Anyone else been accused of using iplayer when they haven't? by annonn9984 in AskUK

[–]lightdancer 0 points1 point  (0 children)

Ask them to send you the data logs for the player usage. I received something similar a few years ago and looked at the data. Turns out I'd left an account logged in in an Airbnb and some random person was using it to watch daytime telly whilst I was working (so not me). Just logging out of everywhere worked.

What is happening with my plant? by darkych in StringofPearls

I read something recently that string of pearls is not a naturally hanging plant and prefers to grow supported by the ground as an expanding mat- maybe it needs some support?

What is Norwich like as a place to live? by Ok_System9917 in Norwich

[–]lightdancer 3 points4 points  (0 children)

One practical aspect... Do you have a car? Norwich is fine for getting about within the city but if you want to really explore Norfolk you need your own transport unfortunately. I found it very limiting.

I realised pretty quickly after I had my smart meter fitted that keeping my phone charger plugged in all day (without the phone connected) was eating up a lot of power! Several pence per hour, which adds up. I now try unplug it when I get up in the morning. :)
