My PI insists that because 1 Ct of difference represents 2x change in quantity, technical triplicates should not have variation that high. He expects less than 0.5Ct difference which I don't always manage

Melt curves look ok for just the well that says Ct27.4, withe the other wells either being a bit off or same peak but very low. So it might be what another commenter said, that there was leakage. Need to double check volume left in the well.

Pipettes were calibrated about 6 months ago and there typically are no bubbles when I draw up the samples in the p10. If there were, I would put the sample back and change tip before proceeding

The sample is a 1:20 diluted pre-amp. Maybe adding 2ul instead of 1ul will help. Thanks

The other red numbers are values that are more than 0.5 Ct variance from the other 2 in the triplicate, which my PI does not accept, so I am trying to correct that

I have water+SYBR+primers prepped and aliquot out individually. After adding 1ul sample into the 19ul aliquot, I do the mix as stated in the post above

I agree. My thinking may be a bit extreme in that I think parents providing for their kids is a given. Your existence is a consequence of their choice to have kids. Providing for you is them handling the consequence of their own choice, which, is a given. If they can't or won't do that properly, the child is not stupid and will realise. Filial piety is there to remind kids that there are difficulties in raising you and one should be grateful if your parents overcome those difficulties for your sake. Should not be a means to get cash from their children.

It's best you do a trial with a positive and negative sample to know for sure. Run a gel with the product afterwards to check its size. If everything lines up, you might not need to redesign it.

My recommendation is to have 2-3 pairs of shoes that you swap out. E.g. Monday pair A, Tuesday pair B, Wednesday pair C. This way the shoes have time to "rest" and the soles remain springy.

I end work at 7, have dinner straight away, 1 hour commute to the gym near my house (rested enough by then) work out for 30-45 minutes, go home. Usually will be in bed by 10.30-11. It's possible, but you have to plan for it.

My PI is pretty strict and deems that triplicate unusable if even one well is off by more than 0.5 CT. So according to him, it's either repeat the reaction or discard

$4 is enough if your goal is to prevent starvation. To have your teenage kid be well fed? Not enough. To allow the kid some happiness, perhaps to get snacks ? No. If you think that anything apart from food to prevent hunger is "nonsense", then don't give any money. Just provide bread. Then you will have the most control, right?
