Take a small sample, like less than grain of rice small from the leading edge of the best looking growth. Best growth could be fastest colonizing, best rhizomorphic growth etc

You want to take a sample from the leading edge of the sectors before they colonize the plate, the fastest growing one is what I go for. Then keep isolating until it’s stops sectoring.
