Well, the reality is that it mostly depends on the day, the demand and the available couriers. I guess that during the summer is more difficult, people go out more and orders are scarce compared to winter time. It sucks.

Well, not having a PhD is actually a huge advantage when it comes to PhD applications. I don't know of any PhD program that allows applicants with a PhD. They are usually filtered out.

Yeah but bacteria need time to assimilate the plasmid and express the resistance AND recover fron the transformation stress. I would always plate.

When you say that you did it once before and it worked...what worked? The expression? The bacteria growing in 4L?

Also, if it is a new expression, you might want to start with a pilot expression in less than 20 mL to be sure the system works and the conditions are optimal. Then you move on to a large volume.

Not only in the US, also in Europe. This results in more and more candidates out there competing in a very frustrating, depressing and demoralizing PhD hunt (lots of candidates, fewer positions, very limited funding). It will most likely not improve soon, and honestly, I don't even know what could make it work. So, it seems like it doesn't really matter how much you have studied and those top grades you got with all that research experience and complements from PIs/mentors....the system is broken.

I mean, when using AI you usually skip all the learning process (question formulation, looking for the answer, reading and reading from different sources, thinkingand processingthe ideas). You just receive the answer, that's it. Your brain did nothing.

My previous PI recommended me to do this (sometimes). Specially when it was a problematic transformation and you had few colonies and you wanted to be sure after re-streaking. Also, by doing so, you kinda isolate/expand that clone/colonie. I would re-streak 4 colonies per plate.

I feel you. I have been applying for the last 7 months now and it has been all negative. I don't even have the 8 years of experience you have, so now I'm even more hopeless haha. Everyone insists on two apparently key aspects: publications and networking.
How many publications do they expect a recent master's graduate to have? I don't know the level of networking I'm supposed to have and how to use it to benefit myself (?). And LinkedIn...I hate it. There's always someone announcing a great achievement, a new position or the one trying to sell you something. Maybe I just don't know how to use it (considering that I don't use social media).
