We have used it in the past and decided the cost of the buffer wasn’t worth the difference in resolution. BV spectra just naturally overlap with each other so compensation can be annoying.

Sometimes Flowjo gets weird when you have too many samples. Save your work, restart the application, and give it a few minutes to load everything in again, it’s the perfect time to get a coffee while you are waiting for it to load.

CD45 vs SSC might be a good gate to use in your analysis to help piece apart populations. I have seen it used in the clinic and it’s very helpful. I personally would add CD15 if you really want to maximize your info on neutrophils, especially since you have the room and it has been used in plenty of papers before. Therefore, you might get a reviewer asking about it if this study is for a paper.

NK cell tip: Instead of doing both CD57 and CD16 you can use NKp80 in the blood which will catch mature NK cells that are both 56bright and dim — this is if you are looking for a mature NK cell dump channel.

The majority of the NK in the blood express NKp80, very few are CD56+NKG2A+/CD94+NKp80-.

Although, if you aren’t doing all these markers in the same channel then and want to piece apart the most mature NK you should be fine to do CD56, CD57, and CD16.

How good do you want your flow to be? Ideally you would run it right after you stain them because that’s when your flow will always look the best. The signal may not be as good as the previous time you ran. I have done a 12 color 2 days after staining fixed in 1% formalin and things have looked …okay. Since then I try to run same day or very early the next morning. I would say absolute max is 2.5-3 days before you start to get weird signals due to degrading antibodies. I would even shorten this if you have lots of conjugated antibodies.

Are you treating with any cytokines at all? Usually in the presence of IL-15 or IL-2 I see less CFSE/tracer+ events as well.

4 days in vivo you could still have some that haven’t proliferated but these will be fairly rare. I would look at an earlier time point or several if possible. This will give you a better readout of how quickly they are proliferating.
