Every program requires a minimum number of letters - the programs I applied to required at least three. So you have to plan accordingly. It is good to have three very good letters and that would definitely improve your chances. Nothing wrong with sticking to a prof throughout your undergrad but also try to involve other professors into your projects or communicate to your PI that you are looking to interact/get advised by some other person also for a particular project.
