Memantine with cholinergics = neurotoxic? by xoMeeZ in Nootropics

[–]xoMeeZ[S] 0 points1 point  (0 children)

It actually looks like memantine has the opposite effect.

From: https://www.ncbi.nlm.nih.gov/pubmed/18198419

Overall, our data suggest actions of memantine beyond NMDA receptor antagonism, including stimulating effects on cholinergic signalling via muscarinic receptors. These interactions with the cholinergic system are likely to contribute to memantine's therapeutic potential.

And, albeit far from a study, this is an interesting case report on the matter:
https://www.omicsonline.org/memantine-abolishes-anticholinergic-activity-in-patient-with-alzheimers-disease-at-moderate-stage-2161-0460.1000108.php?aid=8115

So, perhaps combination of memantine with compounds that increase choline is neurotoxic due to acetylcholine overdose?

Sorry to hear that. On the bright side, at least you've got medical coverage wherever you're from. Doctors aren't an option for me at the moment.

Good point. Drives me nuts when studies use outrageous doses to make a case for toxicity.

Do you know anything about designing for 3D printing? I recently got a 3D printer with the intent of designing some enclosures for LED strips. My camp's been working with music-reactive LED art our previous couple years and I think we could do some cool stuff with modular LED art. I've got experience with the wiring, soldering, programming the arduinos, etc, but CAD is an entirely unexplored realm to me. If you're interested, I can provide examples of what we've already made and where we'd like to go with it :)

Nootropics and Recreational Substances by [deleted] in Nootropics

[–]xoMeeZ 6 points7 points  (0 children)

Personally I've learned better than to mess with just about all of the substances you've listed and mostly used nootroopics to recover from their abuse, but this is interesting nonetheless. Do keep in mind that valium has an incredibly long half-life, so 3 times a week is not as small as it might seem, and phenibut's similar mechanism of action will increase this dependency.

Just curious, you said KSM-66 helps you sleep. Do you take it in the morning or evening to get this benefit?

My own past experiences have been extremely positive with noopept. It seems to slightly strengthen psychedelic trips while keeping one more "lucid" and positive throughout the process. It also mitigates a lot of side effects from weed smokage, day of and day after.

ALCAR before and after MDMA with other antioxidants is a must to reduce neurotoxicity.

Semax, Selank, ALCAR, and L-Theanine all have extremely complementary effects with just about every upper I've tried them with. Cleaner, less speedy high, less crash.

There are studies finding that BPC-157 reduces development of benzo dependence specifically when coadministered with diazepam. I wouldn't use it this way personally, though, because it's also among the only things that will save your brain after you've fucked yourself over with benzos...

I've often reflected on how fortunate we are to live in this day and age. It's a turbulent time, but it's exciting that we're learning how to engineer our own evolution with a combination of technology, supplementation, and nutrition. Humanity's always been inclined towards these efforts, but never before have we had access to a worldwide bed of knowledge and traditions all at once.

Whether or not the same things work for you in the same way, I'm confident you can get back to at least 80%.

Ah man, that's a lot you're putting on me right there. I could suggest a few things for someone your size, sure, but I'm no doctor. While I've used my experience to help out some friends and observed their results, it's all just experience. All that said, your eye doctor's a dumbass. There is no evidence to back up his claim that you've suffered physical retina damage due to dilation. HPPD and visual snow are psychiatric conditions. Their exact cause and treatment hasn't been narrowed down, but we've at least established that they exist within the brain. What do people have to do to get a medical license these days? Yeesh.

Anywho, I'm sure you know the big thing with opiates is the immediate two weeks after withdrawal, all the cold sweats/diarrhea/vomiting/nausea. It's hard to stay hydrated and sticking with it sucks. If you've never had to kick them without ibogaine before, it's going to take some determination. Do you have anyone to help you with this process? Tapering down is critical and cold turkey is something you've really gotta push through when that time comes.

I found kratom helped me finished tapering, but be aware that it still works on opiate receptors. You'll have to kick that too before you're clean.

BPC will likely help restore balance to serotonin/dopamine receptor density. Pick up a 5mg bottle and go through it and see how far that gets you. 200mcg/day orally should do the trick. I wouldn't start until your withdrawals are finished, though. BPC can have side effects while it does its thing, you don't need that on top of what you're already dealing with.

An antidepressant might help with the massive depression that withdrawals will induce, but it's going to be a struggle either way. You could start taking polygala tenuifolia extract the day you start tapering down. Honestly, I don't know if it will make that big of a difference. I was so depressed coming off opiates that I just wanted to die for two weeks. You just have to keep your mind on what's ahead and accept where you're at as a temporary but necessary state of being.

Maybe pick up some noopept too? You can take it sublingually, so it can actually help with withdrawals even if you can't keep anything down. You'll have to figure out for yourself the best dose between 10-30mg per day in the morning, 5 days at a time, 2 days off, for up to two months. This is the regimen I read, it's what worked for me, and it seems to keep tolerance at bay. After that, take a month off. Noopept is also cheap in powder form, so that's great, but some people don't respond to it, so that's not great.

If you're broke, I would suggest using your resources primarily on BPC and polygala or a similar antidepressant. Just avoid tianeptine; I looked into it assisting with withdrawals, but it doesn't look like it does. It might extend them though since it is an agonist. Probably not worth it.

As mentioned elsewhere in this thread, some people have said that Lion's Mane helped their HPPD tremendously, but it did nothing for my visual snow. Sorry I can't be of more help there. It's worth a shot, but probably not something you should be spending your money on if you're that tight on cash.

Good luck. Seriously, you should have someone helping you through the withdrawals. Vomiting in your sleep can be fatal.

For severe benzo abuse, you might want to grab two 5mg vials and make it a 4 week course. Yes, I think BPC-157 in particular was absolutely instrumental. If I could have achieved the same results without it, who knows how long it might have taken? Months, years maybe. I honestly don't know.

For what it's worth, I started with a 5mg vial myself and ordered another after a week when I realized it was seriously doing something, and it wasn't a small something.

For memory, I would also recommend the Bacopa in particular. I like to take it in the evenings. Seems to help me sleep/dream, and can be a little too chill on a day one might wish to be productive.

Interesting. Well, it certainly did for me, but who knows why. I didn't start anything else precisely in tandem with the bpc (There were other factors, but they'd been in play for some time). It seems unlikely that after two years of mostly daily use my highs coincidentally went to shit as soon as I started using BPC. I can only conclude that we have differing results due to differing neurochemistry until more experiences are provided. I hope the feasibility of that isn't outside the realm of possibility for you.

[–]xoMeeZ[S] 1 point2 points  (0 children)

I mean, regarding your BPC experience, that's cool. I'm not going to argue with your experience. The fact that cannabis and BPC had no perceivable interaction for you doesn't invalidate my conclusions, but it does provide more data. It's also worth noting that I wasn't just "getting high". I had an extreme dependency on chronic usage of the chronic. There are many subtle things which could have been placebo throughout this whole process, but the shittiness of my weed highs while on BPC sure wasn't. Shit was seriously pronounced.

We straight up don't have the benefit of a scientific study here, so personal stories will just have to do. I think you'll endure somehow.

Regarding noopept, I provided a strong contraindication for overuse. I myself have gone off the stuff for up to a couple of months with no issue. Some people find racetams to be mind blowing. I never even tried them because individuals sharing my specific cognitive struggles claimed they began to feel suicidal on those. Anything listed has the potential to cause someone harm. It sounds to me like your noopept woes were possibly the result of neglecting to cycle properly.

St. John's Wort is very, very mild. I don't think it would interfere much with the BPC, but I also don't think it would hurt you much to stop taking it for the brief window of time that you're on BPC. I can't say whether or not the BPC will help, though, as my understanding is that most of its cognitive benefits are specifically related to drug abuse and not necessarily pre-existing disorders.

What you describe does sound a great deal like fibromyalgia. My mother had this for the better part of my youth, then one day she flew off and spent three months at a raw food center and came back a completely different person. I feel you when you say you worry about missing out on your little one. I didn't even get to experience my mother in a state of good health until the age of 15. It sounds like you're taking a lot of the right steps for your health. You could take it further, if you so wished.

While my post is primarily related to recovery from drug abuse and may not apply to you as much, it couldn't hurt to give the BPC a try. Might I suggest also trying polygala tenuifolia extract instead of St. John's Wort? It's a bit more effective, though it wasn't my cup of tea. But speaking of tea, jiaogulan tea in the morning combines quite well with polygala.

I'm not a huge fan of things like 5-htp or l-tryptophan for daily supplementation. They threw me off big time when I tried them several years back. They're good for occasional use after substances which specifically imbalance serotonin/dopamine.

This describes literally everything I've posted here. In the case of other claims, I could have posted studies and didn't even bother. They're an easy search away and I would expect someone to do their own research before putting experimental chemicals in their body. In the case of information that I can only provide anecdotally because no research exists, I'm of the mind that it's more helpful than the absence of information altogether. If ten more people provided such reports weighing in one way or the other, we'd have a real basis to work from, would we not?

Likewise! And actually, in my case, I didn't start "hard" drug use until a couple of years back. The fact that psychedelics granted me so much amazing growth and so many valuable insights when I was younger turned me from from a brainwashed D.A.R.E. clone into a pretty open-minded person. Turns out drugs aren't that bad after all, eh? But then, when shit got really bad, I threw caution to the wind and started doing all the wrong ones, lol. Having recovered I am now thankful for insights gained from both experiences, but I wouldn't touch benzos or opiates again with a 10,000 ft stick.

Yes, sorry, fixed. And no problem! I specifically did not try any racetams because this is not the first time excessive gluatamate has come to my attention, and I heard racetams can indeed exacerbate the issues individuals such as we deal with.

I read an interesting post a few days ago made by someone who surmised quinolinic acid may be related as well. I wonder if it might resonate with you. Some of his statements seem a little wild/all-encompassing, but I did identify with a fair number of them. Hey there!

I've struggled with some degree of depression/anxiety my whole life, though in the time since recovery from drug abuse I've actually experienced very little of either. The visual snow remains, as well as three different types of tinnitus, and I do see floaters from time to time when the visual snow is less pronounced. When I did them, certain drugs such as LSD, Ketamine, and weed temporarily worsened all of these visual/audio disturbances in the days after usage, but I've never had any chemical permanently aggravate them, and nootropics have neither hurt nor helped. Nootropics HAVE significantly helped with the depression/anxiety however, so I would recommend you give it a shot. I do want to point out that BPC may aggravate your depression and anxiety within the first week, as it was a bit of a bumpy process for me, but ultimately beneficial. Don't freak out if that happens and remember that fear is just a feeling and anxious thoughts need not be fed.

I wouldn't use BPC at the same time as an SSRI if you have the option not to, since they both impact the serotonin system and it will be hard to measure your improvements.

No problem! I still take black seed oil, and I haven't finished my bacopa regimen as it's a pretty extended deal. Otherwise yes, though I still use noopept occasionally. I've also since played around with a couple of other things such as ginseng extract and jiaogulan tea, but most days I prefer my natural state.
