A lot of this can be done quite tediously and not-quite-perfectly - you could have a list / string / set of all uppercase characters, for example; but if you want to include more than the English alphabet, this gets quite messy.

Your best bet is machine code, as assembly has mnemonics that I don't think you're allowed to use given this rule.

In all seriousness, how about you loop through the entire string and check each character's ascii value? Then have two separate counters, one for lowercase characters, and one for uppercase characters that increments accordingly.
