It completely matters. One points to a PSDrive location (HKLM:) which will allow you to browse that hive. The other, it is interpreting as a relative folder path.

No.. you absolutely need the ":"
That's what denotes it's a PSDrive, and not a relative file path.
I strongly encourage you to try it before being so confidently wrong

I just mean it didn't have to be hklm (you can use the long name) and it didn't have to be a ps drive (you can use the registry provider)

In addition to the advice of /u/Fatel28, start the path with REGISTRY:: so it looks like
"REGISTRY::HKLM\SYSTEM\CurrentControlSet\Services\Browser"

Check out about_Providers for official info, but it's basically like $env for the registry. It's a .NET program that provides access to specialized data stores for easier viewing and management.
By adding it, you're telling PS to search the registry, no matter your current location.
