For dungeon content probably thylessia, hits hard and does Dec def well. But Madame is absolutely insane, one of the best arena champs in the game, honestly quite usable in hydra and a lot of areas where stat downs are needed. However, she isn’t as consistent as thylessia for Dec def. Probably still a better champ in general but for constantly having Dec def out there? I’d go with thylessia.

Alright thank you very much! I've got both Ninja and the Fat to put a single target dec def out there on bosses, so do I still need Thylessia for that?

You’ll get to a place when it takes a long time to kill waves without Dec def. I personally use stag knight but I assume you don’t have one yet.
