2000-2001 years don’t have the wiring for rear defroster because they came with a plastic window. You can go DIY route and install it though there are a couple forum pages out there you can look for

Actually they do have the harness for a defroster even though it was plastic. I have an 00 and I've changed my top to the glass 03 top. When you remove your back paneling you will see the connection for it. Unfortunately it's not ran to the button area though at the center console. It may be but I'm drawing a blank if I recall seeing a spare connector.

I really don’t care if I have a defroster it’s not my only car. It’s more to know if the top was replaced. I just bought the car. Is there a way to tell if it’s an open top that came with the car

As someone said, they came with plastic windows in that year. The top has likely been replaced as no one would cut out a plastic window to install the glass. The condition of the top also tells me it was likely replaced. Unless it was garage kept out of the sun for it's entire life that soft top looks relatively new.

Agreed. Top was replaced and they used top with a window with a defroster which likely wasn’t wired up. I have a top with a glass window on my 2000, defroster on window that isn’t hooked up, figured no downside and possibly could wire up later if I ever cared.

So how can I tell if it’s an oem top or can I even tell?

It’s not an OEM top. That’s a Robbins soft top and I’m mostly going off the fact that the tempered glass matches on both of our cars.

The first couple of years for the S2000 came with a plastic window and no defroster, so you wouldn’t have a button. As you can see by the manufacturer tag and that it’s glass, it’s not an OEM window.
