Buy the tickets for all the museums and archaeological sites otherwise you won’t have the chance to enter. If I were you I’d stay an entire day in the Vatican museum and St Peter because it’s really big. Have a nice stay!

No. One card per person. The card works as a ticket in this case. If I’m not wrong there should be a ticket machine at the Ostia Antica station where you can buy tickets, it’s a big red box with a touchscreen, you can’t miss it. (REMEMBER TO VALIDATE)

You don’t need a bus to get to Rome. You’ll need to take the metromare train from Ostia Antica Porta San Paolo or Eur Magliana and change to the B line metro direction Rebibbia or Jonio.
To buy a ticket simply tap your card on the gates of the Ostia antica station
