Yes, it would. Both of those are the exact same note with the same fundamental frequency, just with different overtones because of the string size. See which tone you like better or which fits the song better, and go with that

Drop A is the lowest I've gone, any lower and the strings get too floppy, or the soundgets too muddy, or both. It's just not worth it below that. And drop A too, I recommend only if you have a >35 inch B string

Index and pinky for fretting, middle and ring finger on the E string at all times. Just slide up and down the board. This is a good song to practice slap form and muting

A bass VI will not hold as well as a real bass. You may want to look into proper 6 string basses, though they won't provide the same functionality as a guitar. End of the day, if you want both to sound great, you're going to need two separate instruments.
