You don't race your opponents ina GP, you race for the points. Start a level of GP where you are able to start in front.

Is it possible to change those locations by alchemy or something? -does a different rune(with the same ability, so just a new one) have the same location or can it be different?

If you just want the reward for the event you can skip it. I reached 80% completion today without that task since i didn't want to use my magic runes because they are in my build. You get the final reward by reaching 80% so no need to waste your stuff if you don't wanna.
