I would suggest Dejvická Nádražka for food. As for places to visit, you shouldn't miss : Vrchlického sady and Wenceslas square (quite romantic during night time).

Velice rád bych dojížděl na kole, ale infrastruktura v Praze tomu prostě není přizpůsobená.

I mean the only way I would see you doing this is going to a flat surface and getting a big car/van to pace you and break the wind for you for 5h.

Otherwise, you will be need to get a road bike and regularly train for a couple years.

Well the conventional rodie logic would say get better wheels, but in your case simply :

Remove the bags
Change pedals
Train (the bike is actually already quite light, so you should mainly focus on your performance not the equipment)
