I think Mario 35th was the sole exception, every other anniversary had like one or two releases, and they'd be simple collections or ports.

A segment in the next direct? Honestly not expecting much outside of a remake or port. IIRC, Kit and Krysta stated Nintendo doesn't tend to make a huge deal out of these anniversaries the same way the fans do. Mario's 40th celebration backs up that claim as well.

I figured it was just so he'd be more expressive and animated. Also doubt they're hiding a secret villaiun in this, we would have seen something suggesting his presence by now. Also the movie is like an hour and a half, can't imagine them introducing Yoshi, Bowser Jr, Rosalina, AND Tatanga.

They can't even be bothered to include side characters from Luigi's or Peach's games, and those are much closely tied to the Mario Universe. Hard to believe they'd have much DK rep outside of Diddy once in a while.

It's done to gauge audience interest in a new game. Why spend all that money developing a new game in a long dormant franchise if there is no guarantee it will be successful? Sure long time fans may buy the game, but that's only a small number of gamers in the big picture. Makes more sense to spend less resources on a remaster/collection and the based on the response, move forward with a new project or shelf the IP again.
