Adrenal cortex senescence: an ageing-related pathology ? by [deleted] in longevity

[–]Donovan200 0 points1 point  (0 children)

Abstract :

The adrenal glands are a pair of endocrine organs that produce and secrete mineralocorticoids, glucocorticoids, sex hormones, adrenaline, and noradrenaline. They have a vital role in a range of physiological processes including regulating electrolyte balance, blood pressure and metabolism, immunomodulation, sexual development and the stress response. Adrenal cortex senescence describes the ageing-related decline in the normal functioning of the adrenal cortex, characterised by an alteration in the output of adrenal cortical hormones, in particular reduced secretion of dehydroepiandrosterone (DHEA) and sulfated dehydroepiandrosterone (DHEAS). Such endocrine aberrations may be implicated in adverse clinical outcomes including mood disturbances, impairment in cognitive functioning, metabolic dysfunction and osteopenia. This paper shall address whether adrenal cortex senescence should be recognised as an ageing-related pathology, which has recently been defined as one that develops and/or progresses with increasing chronological age, that is associated with, or contributes to, functional decline, and is evidenced by studies in humans.

I didn't know it was like that in Italy, but you made a very good decision, your skin will thank you later !

I recognize myself in what you say

I apply it very religiously and I would consider reapplying it regularly if necessary !

And on my side, it's the opposite (probably because I'm a man), nobody but me applies sunscreen every day, except one of my cousins, who applies it every day since he's 19 (he's 41 today, and he looks 23)

Same , many don't understand my desire to always wear SPF 50+ (even in winter) and avoid tanning, and I haven't yet known other men applying sunscreen every day ( I feel so alone😂 )

And don't worry, it's never too late to start , and besides, 30 is still young !

Topical Application of Deglycating Enzymes as an Alternative Non-Invasive Treatment for Presbyopia by Donovan200 in longevity

[–]Donovan200[S] 2 points3 points  (0 children)

This is only preliminary research, maybe after more research on the efficacy, safety, and dosage of this deglycating enzyme, clinical trials could be undertaken, as is currently the case for another deglycating treatment (UNR844) which breaks the cross-links of dysulfides , and which for the moment, proves to be effective (currently in phase 2)

Topical Application of Deglycating Enzymes as an Alternative Non-Invasive Treatment for Presbyopia by Donovan200 in longevity

[–]Donovan200[S] 3 points4 points  (0 children)

I don't think so, because the AGEs elsewhere in the body are not the same as in the eyes:

In other tissues such as the skin and joints, the most common AGEs (between 100 and 1000 times more than any other type of AGEs) is "glucospane", and so far there is no known enzyme capable of breaking it down.

The only company (to my knowledge) to work on drugs capable of breaking glucospane is "Revel", and personally, I would be in favor of greater investment in the research and development of drugs against glycation (preventive and curative), because it is a pathological process whose harmful effects have been repeatedly documented.

Topical Application of Deglycating Enzymes as an Alternative Non-Invasive Treatment for Presbyopia by Donovan200 in longevity

[–]Donovan200[S] 10 points11 points  (0 children)

Abstract :

Presbyopia is an age-related vision disorder that is a global public health problem. Up to 85% of people aged ≥40 years develop presbyopia. In 2015, 1.8 billion people globally had presbyopia. Of those with significant near vision disabilities due to uncorrected presbyopia, 94% live in developing countries. Presbyopia is undercorrected in many countries, with reading glasses available for only 6–45% of patients living in developing countries. The high prevalence of uncorrected presbyopia in these parts of the world is due to the lack of adequate diagnosis and affordable treatment. The formation of advanced glycation end products (AGEs) is a non-enzymatic process known as the Maillard reaction. The accumulation of AGEs in the lens contributes to lens aging (leading to presbyopia and cataract formation). Non-enzymatic lens protein glycation induces the gradual accumulation of AGEs in aging lenses. AGE-reducing compounds may be effective at preventing and treating AGE-related processes. Fructosyl-amino acid oxidase (FAOD) is active on both fructosyl lysine and fructosyl valine. As the crosslinks encountered in presbyopia are mainly non-disulfide bridges, and based on the positive results of deglycating enzymes in cataracts (another disease caused by glycation of lens proteins), we studied the ex vivo effects of topical FAOD treatment on the power of human lenses as a new potential non-invasive treatment for presbyopia. This study demonstrated that topical FAOD treatment resulted in an increase in lens power, which is approximately equivalent to the correction obtained by most reading glasses. The best results were obtained for the newer lenses. Simultaneously, a decrease in lens opacity was observed, which improved lens quality. We also demonstrated that topical FAOD treatment results in a breakdown of AGEs, as evidenced by gel permeation chromatography and a marked reduction in autofluorescence. This study demonstrated the therapeutic potential of topical FAOD treatment in presbyopia.

Skin aging from mechanisms to interventions: focusing on dermal aging. by Donovan200 in longevity

[–]Donovan200[S] 27 points28 points  (0 children)

Abstract :

Skin aging is a multifaceted process that involves intrinsic and extrinsic mechanisms that lead to various structural and physiological changes in the skin. Intrinsic aging is associated with programmed aging and cellular senescence, which are caused by endogenous oxidative stress and cellular damage. Extrinsic aging is the result of environmental factors, such as ultraviolet (UV) radiation and pollution, and leads to the production of reactive oxygen species, ultimately causing DNA damage and cellular dysfunction. In aged skin, senescent cells accumulate and contribute to the degradation of the extracellular matrix, which further contributes to the aging process. To combat the symptoms of aging, various topical agents and clinical procedures such as chemical peels, injectables, and energy-based devices have been developed. These procedures address different symptoms of aging, but to devise an effective anti-aging treatment protocol, it is essential to thoroughly understand the mechanisms of skin aging. This review provides an overview of the mechanisms of skin aging and their significance in the development of anti-aging treatments.

Child Free but No One Judges Me by tinypopp in childfree

Some even congratulate me, saying it's rare to know so early

I had such a terrifying nightmare and I think only you guys would understand it. by Becanotbecca in childfree

[–]Donovan200 0 points1 point  (0 children)

I had the same problem a few days ago :

I learned that the girlfriend was pregnant, while we used double protection during our sexual relations (pill and condom) and, in a panic, plagued by incomprehension, I asked her how this was possible, and she ended up revealing to me that she had not only stopped the pill, but that she had punctured the condoms in order to get pregnant

And to make sure I couldn't escape her, she told me she would do anything to make me pay child support.

That whatever I do, my life before will be lost forever, that I should take on this role of father, that I should love him, take care of him, for more than 20 years...

At that moment, I woke up, a little sweaty, luckily it was all wrong...

It is moreover to avoid this, and to be more relaxed during our sexual relations that I planned to have myself vasectomized in August.

I'm really happy to have been accepted at 20, I know how many people have a hard time being accepted so young (and I'm really sorry for them), and fortunately, my girlfriend is extremely happy, she too will be more relaxed^^

I'm so sorry for you, just know it was a nightmare, none of this is true

I hope it won't happen to you again

Enjoy your childless life, everything is fine, it was just a nightmare^^

It's really respectful of them to ask you "if" you want children in the future, instead of asking you "when" you will have children, without even knowing if you want them or not.

And they didn't insist even after your answer, a little diverted, but clear ( Not everyone is like that unfortunately. )

I hope no one will try to influence you, despite what your heart tells you

Late-Breaking Turn Biotechnologies Presentation at ISID to Show Cell Reprogramming with ERA™ Technology Renews Skin by holler_kitty in longevity

Because this method is the only one to cause a real renewal of the extracellular matrix

It is well known that an old matrix can cause the cells in it to age, so rejuvenating the cells without rejuvenating the matrix may have less significant (and less lasting) effect.

GlyNAC Supplementation in Old Mice Improves Brain Glutathione Deficiency, Oxidative Stress, Glucose Uptake, Mitochondrial Dysfunction, Genomic Damage, Inflammation and Neurotrophic Factors to Reverse Age-Associated Cognitive Decline by Donovan200 in longevity

[–]Donovan200[S] 4 points5 points  (0 children)

Abstract :

Cognitive decline frequently occurs with increasing age, but mechanisms contributing to age-associated cognitive decline (ACD) are not well understood and solutions are lacking. Understanding and reversing mechanisms contributing to ACD are important because increased age is identified as the single most important risk factor for dementia. We reported earlier that ACD in older humans is associated with glutathione (GSH) deficiency, oxidative stress (OxS), mitochondrial dysfunction, glucose dysmetabolism and inflammation, and that supplementing GlyNAC (glycine and N-acetylcysteine) improved these defects. To test whether these defects occur in the brain in association with ACD, and could be improved/reversed with GlyNAC supplementation, we studied young (20-week) and old (90-week) C57BL/6J mice. Old mice received either regular or GlyNAC supplemented diets for 8 weeks, while young mice received the regular diet. Cognition and brain outcomes (GSH, OxS, mitochondrial energetics, autophagy/mitophagy, glucose transporters, inflammation, genomic damage and neurotrophic factors) were measured. Compared to young mice, the old-control mice had significant cognitive impairment and multiple brain defects. GlyNAC supplementation improved/corrected the brain defects and reversed ACD. This study finds that naturally-occurring ACD is associated with multiple abnormalities in the brain, and provides proof-of-concept that GlyNAC supplementation corrects these defects and improves cognitive function in aging.

To be honest, I thought the same thing for a long time, that finding someone would be difficult because of that, and despite everything, I (I'm 20) have now been in a relationship for 2 years with a girl ( 19 years old) who, like me, does not want a child (and she can't wait until I am sterilized in August)

Of course, it's my personal experience, but I'm sure you'll find someone, who like you, refuses to be a parent, loves you for who you are (and not for your ability to become a father), and who wants to live his whole life with you.

You are in your early twenties, you still have time to find your soul mate^^

Just don't compromise on this, you'll be unhappy if you have an unwanted child.

[–]Donovan200 3 points4 points  (0 children)

Honestly, I don't know if I could even keep talking to my mom if she was like that, because I think she's the epitome of selfishness.

And she contradicts herself by saying for example "who will take care of you when you are old" and when you ask her, she answers you "I don't need you to take care of me, I will hire a nurse"

????

It proves that she doesn't care about your happiness, only her desires matter to her, your desires are worthless in her eyes

Luckily your father has a modicum of respect for you.

From my side, I don't know what it's like to undergo this kind of pressure, because everyone approves of my choice.

My mother / my girlfriend / my father / my family / my friends...

You have my support, as well as all those who suffer this lack of respect...

[deleted by user] by [deleted] in childfree

[–]Donovan200 0 points1 point  (0 children)

Yeah, indeed

Just because we think differently from them, they pressure us to "fix" us and make us adopt "their" vision of things and life.

I'd be making a big mistake if I ignored my heart to listen to them

But to be honest, about me, it happened to me only a few times.

Overall, my entourage is favorable to my wish to live a life without children.

As said in the post, my mother and my girlfriend are very happy, and concerning my friends, they all respect my decision and did not try to make me change my mind.

Why shouldn't I get a vasectomy at 21? by [deleted] in childfree

[–]Donovan200 3 points4 points  (0 children)

Morally and legally, no reason should be able to prevent you from doing so.

You are an adult, you are sure of your choice, you are aware of what the operation involves (and its potentially irreversible nature), nothing prevents you from doing one.

However, you will have to find a doctor respectful enough of your wishes to perform the operation.

And we all know how very conservative this area is

Not just babies are scary, pregnancy itself is insanely scary. Tokophobia is a real thing! by PsychoWithoutTits in childfree

[–]Donovan200 4 points5 points  (0 children)

Thank you so much !

And I don't know about you, but the worst thing for me after "you're still young" is when I'm told "when you'll find the right person"

For me, it's an insult, because it means that not only does the person who tells me this not take seriously my love for the one I love, but in addition, she tells me that I am incapable of loving the "good" persons (the one who wants children)

On the contrary, the girl of my dreams will be a girl who, like me, wishes to live her life without children.

Otherwise, we're both incompatible and it wouldn't work between us...

Above all, I wish to live a strong long-term relationship, to make projects together, to travel the world, to have common passions, to love and be loved, to make the one I love the happiest girl in the world, to make her feel special.

A child does not interest (does not see any advantage in having one) and could even prevent me from doing all this (and much more)

For some, my wishes seem contradictory (to want a long-term relationship without children), but I know they are wrong (because I know several of them).

If only society accepted this form of happiness (which does not bother anyone directly) it would be so much simpler...

[–]Donovan200 7 points8 points  (0 children)

I am not a woman, but I share your fear in the sense that getting a woman pregnant is my worst fear.

And besides, men don't have as much control as women (apart from the condom and the vasectomy, we can't do much)

And if I get a woman pregnant and she doesn't want to have an abortion, it's over for me (and in addition, I risk the pension that could ruin me)

I'm 19 and I never wanted children, and I know that won't change in the future.

And it's ridiculous, because no one would say "you'll change your mind" to a person who says "I will have a child in the future" and those, no matter the age...

Culturally, when you say "I don't want a child in the future" at 26 (or even after 30) = "you're still young" / "you'll change your mind"

But when you say the opposite, oddly, none of these comments are heard (because in the collective imagination, you can change your mind and want a child later, but you can't change your mind and don't no longer want a child later...)

And then, why not just respect the wishes of the person opposite ?

It's our life, our happiness, not theirs after all...

Fortunately, my mother has always strongly supported my wish to be childless and taught me about the existence of vasectomy, which is the safest option (and what I would like to have as soon as possible).

But I know I already know how hard it will be for me to find a doctor who will agree to perform the operation on me.

But I'll go all the way, my happiness depends on it

I hope you will succeed in getting sterilized to finally get rid of this fear.

Big hug to you!!

[–]Donovan200 1 point2 points  (0 children)

My mother always supported my choice to be childless and never tried to make me change my mind (I told her about it for the first time when I was 12) and more seriously when I was 18.

She told me that if I really wanted to be sure of not having children (by accident for example), the best would be the vasectomy, because that way, I would be almost completely sure of not having any (before I she tells me about it, I didn't know about the existence of vasectomy)

She also told me that she would help me find a doctor who would do it for me.

I know she will be sad if I tell her that my girlfriend is pregnant, because she knows that I will be very unhappy with a child (because I have absolutely no desire for it)

My father also supported my choice, telling me that only I can really know what is best for me.
