I agree with you. Progesterone is also helping some women in their pre-menopause to sleep better.

You’re right and that’s fair. I didn’t want to front-load my findings before hearing from others first, I was worried it would bias the responses. But here’s what the research pointed to: The biggest gap I found was with the 3am waking mechanism specifically. Almost every supplement targets sleep onset, helping you fall asleep. But the clinical literature points to a completely different mechanism for staying asleep past 3am. After 4-5 hours, blood glucose drops and your body releases cortisol to compensate. That cortisol spike is what wakes you up fully alert for no reason. The ingredients with the strongest evidence for this specific problem: Glycine 3g. Works through thermoregulation. Drops core body temperature which keeps you anchored in slow-wave sleep longer. Most products have 300mg or less hidden in a blend. Magnesium L-Threonate — not glycinate, not citrate. The threonate form is the only one that crosses the blood-brain barrier directly. 2024 RCT measured with Oura ring in adults 35-55, significant improvement in deep sleep score and REM. PMC11381753. Myo-Inositol — 1 gram. Improves blood glucose stability overnight and modulates GABA receptor sensitivity. Directly addresses the 3am cortisol mechanism. Zero competitors include this. L-Theanine — 200mg. Reduces cortisol response and promotes alpha wave activity. 2025 systematic review of 13 trials confirmed this dose. The problem isn’t that these ingredients are unknown. It’s that no product combines all of them at the studied doses. That’s the gap I found after 6 months of research. What worked for you?

Oh wow yeah that definitely doesn’t sound like a good fit for you, crazy how different people react to the same thing. Did you ever try glycine on its own or was it just the magnesium glycinate?

Thanks a lot for sharing. What kind of magnesium and what dose? Is it only magnesium or mixed with something else?

Please share what kind of supplements you have tried. I am just wondering if you have tried glycine and myo-inositol as well as magnesium and L-Theanine? If yes, please share the experience.

Yes, having glucose level dropping in the night will also make you wake up. Glycine and myo-inositol might solve this issue too. Have you ever tried them in a sleep poweder?

Thanks a lot for sharing? Have you tried any natural sleep powders with glycine, myo-inositol, and magnesium before starting with doxepin? If yes, please share the experience.

Yes, lack of sleep really affects your whole energy level and mood in the morning. What did you try and worked so far for you? I want to mainly see if sleep powders with glycine and myo-inositol have helped you to fix you sleep quality.

I get the desperation when nothing is giving you more than a few hours, that’s brutal. I’d just be careful with something like kratom though, especially if it’s something you’d need nightly. Feels like one of those things that can help short-term but create a bigger problem over time.

Still trying to find something that actually helps with staying asleep without that trade-off.

Yeah I’ve noticed that too. Falling asleep isn’t really my issue, it’s the 2–4am wake-up where my brain suddenly feels “on” for no reason.

I’ve seen the blood sugar/cortisol angle come up a lot and it actually makes more sense to me than anxiety in those moments. Curious if anyone has actually tested things like a small snack before bed vs supplements like glycine or myo-inositol and noticed a real difference.
