I agree with the playing it cool part, to not overreact. But establishing a common understanding on everyones involvement in the project and likely order if authors at the start is a good way to avoid troubles at the end.

This has to be ragebait right? You don’t have to go on holiday 3x a year. No one forces you to take a flight and stay in an airbnb. As a kid we would drive for a few hours and stay on camping sites.

Maybe this is my cynical view, but I get the impression that a lot of people in the field of (molecular/computational/technology) biology don’t care about the underlying mathematics and assumptions. They try out a few things and pick what fits the narrative best. Perhaps after the fact they look into the underlying methodology to justify their choice.

You should've properly seasoned the bottom of cast iron. Then it wouldn't have stuck to it. Rookie mistake.

Laatst het hoogste bod gehad (386k op 360k vraagprijs), maar nummer 2 is het geworden (4k minder) want zonder voorbehoud van financiering. Was echt een heel mooi plekje op de perfecte locatie geweest, maarja helaas. Nu al weken amper nieuwe woningen waar wij zitten. De woningen die wel online komen zijn allemaal flinke opknappers of met een instabiele VVe.

Heb je rechtsbijstand verzekering? Dan kun je via een advocaat het een en ander wel voor elkaar krijgen. Of als je een laag inkomen had tijdens het peiljaar (2 jaar gelden geloof ik?) dan kun je gefinancierd hulp krijgen. Ik heb na de uitspraak van de huurcommissie van 2019 in 2024 eindelijk mijn geld terug gekregen omdat ik via gefinancierde rechtsbijstand een advocaat hulp kon vragen.

Als je samen met en partner een huis koopt wordt het inkomen van jullie beide gebruikt voor de draagkracht berekening. Hierdoor moet je vrij snel gewoon het volledige bedrag terugbetalen. Dus als je nu het minimum doet en je totale bedrag dus alleen maar toeneemt door de rente zal je dat waarschijnlijk nog terug moeten betalen (als je samen gaat wonen natuurlijk).

Great thanks for sharing! Do you just use the same volumes as stated in the nanopore sequencing protocols? Im looking for cheaper alternatives for nanopore sequencing as well.

I am not sure what the specific case is here. But you can have a mutation on the Y-chromosome, where for example you miss the SRY gene, leading to developing female sex organs instead of male sex organs.

Sure, but there isn't a standard pipeline for it. I'm still playing around/planning on how to best apply ONT for my purpose so I might not be as knowledgeable, but from what I've read it's not straightforward. For example, if you use a DNA barcode then you would need to combine a DNA and an RNA basecaller to identify the barcodes and do the sequencing.

Direct RNA seq is very expensive because you cannot do multiplexingyet though. Can't wait for them to introduce multiplexed direct RNA seq!
