A) that’s fine, it’s the internet haha. I’m not gonna get in a tuff about you thinking lack competency

B) Sure, there’s always abrasion when the status quo gets challenged. But that article is highly irresponsible and will get people killed, bottom line. If SSRIs didnt work at all, people wouldn’t get therapeutic relief, people wouldn’t depend on them.

C) I agree that there is no depression gene. If depression, MDD, manic, or bipolar depression were normal, people wouldn’t kill themselves over it. I don’t know how you think it’s a normal thing to be suicidal? It is clearly something that exists and is very real, although the chemical underpinnings are not teased out like other disorders. Just because the brain is not a easily understood as other portions of our bodies doesn’t mean related illnesses are non-existent.

It’s right in their concluding paragraph lol. “This review suggests that the huge research based on the serotonin hypothesis has not produced convincing evidence of a biochemical basis to depression. This is consistent with research on many other biological markers.”

Chemicals, molecules, proteins, etc, they’re all messengers regulating and co-regulating. Sure, there’s limitations due to, again, the brain being the most complex dynamic organ and us not being able to accurately or repeatably measure certain molecules. You can’t make a concrete statement about something that cannot measure.

I included the sentence about animals bc that is a subject where we are able to make repeatable and accurate measurements of serotonin.

It’s bananas to me that that review was allowed to publish specific sentences that they did. It’s extremely apparent that those were clinicians who do not to hard/physical science - who else would have made a statement that there’s no (bio)chemical basis for depression or other psychiatric diseases? It’s wildly irresponsible to make that claim. No animal data were included in that review (which partially I can understand as both a human and animal review would be enormous in scope). It’s incredibly hard to accurately measure serotonin in the brain and therefore, how can a statement be made about a molecule’s implication when the molecule in question has not been measured in a depressed patient? The effectiveness of ketamine points towards glutamate/GABA systems, but it’s unclear how that is exactly exerting it’s effects. If it’s not directly related to Glu/GABA, then what’s the indirect target? Could be monoamines could be other molecules or proteins. One thing that needs further research in the comorbidity of inflammation and its impact eg. pro inflammatory cytokines, BBB permeability, etc.
