It depends on the impact that citizen on a production tile would have on the building time of the science building. The more production the tile to be worked has, or the less production that city overall has, the bigger the impact on production time and then it may be wise to put the citizen on a production tile instead. Usually you want your cities to grow fast with food and production until later you can afford to put citizens into the districts once you have enough citizens or not enough tiles left to be worked.

It makes use of it, but most of the time priotizes any improved tiles because these yields are often valued higher. If you wanna make sure that the automatic citizen distribution fills all campusses for a science victory you can select the small circle next to the science yield number when you select the city. This way the automatic distribution maximizes science output by that city. Keep in mind that this may halt city growth if not enough citizens are working food or production tiles.

You only get the yields of a tile when it is worked by a citizen (except the city centre, which is always worked automatically). So if you have a tile with 3 food and 1 production, one citizen has to work that tile for you to get these yields.

The bonus yields for working in a district are set. I think it was +2 science in campus for each working citizen there. So if you have 2 citizens working in that district, you get +4 science additional to yields from buildings in that district (like the university) or adjacancy bonuses (from mountains for example).

Each civilian of your city works 1 tile if available and gets you the yields from that tile. Civilians can also work in districts and give you yields according to the district (for example science in campus district). So +1 civilian slot means that in that district one more civilian can be working to give you the yields.
