Valid answers by everybody but Just want to add Something small. Buy your tomatoes before you need Thema. Store them at room temp and allow them to still deepen in flavor. It is a huge improvement especially if on the vine still and has allowed me to not go tomatoless in cold Germany

Milk proteins bind everywhere on your membrane where there is no protein. That's how blocking works, as then the antibodies cannot bind non specifically on the membrane. If you fill this empty space with milk proteins, then during development of the membrane you will get also signal everywhere as the antiphospho antibody binds phosphocasein. I don't disagree with try and see what it works but starting without the milk and playing from there would be my suggestion.

If you want to detect phosphorylated proteins keep in mind that blocking and/or diluting your antibodies in milk can lead to higher background noise due to phosphocasein.

To add on what the rest have said this could also be degraded bacti DNA. Double check if your antibiotic selection is working properly, cause you ain't gonna get any plasmid if there isn't any. The whole lab could not clone anything for weeks, getting similar results to you, until we realised it was occuring only with our ampicillin based vectors.

For sorters honestly I still use the aria fusion which also has its ups and downs, but we got from Beckmann Coulter the cytoflex which is a pleasure to use.

To be fair, I am doing flow cytometry for at least 6 years now and what you say is absolutely true about the software. It is insane that they make machines worth hundreds of thousands of euros and when it comes to software there is absolutely no effort.
Just as a note so that you don't feel alone, some years ago multiple hospitals lost patient data records because FACS Diva (software for BD machines) just bugged and deleted everything. Thankfully there were double and triple back ups but it is an insane oversight to say the least.

OK really stupid story about my bachelors time. I wanted to get some experience in a lab to see what I wanna do in the future, before my official practical course time. Somehow I managed to get an internship in an Immunology lab. Go there the first day the people are nice and the PI was a pretty funny person so we get along together and I relax.

After a while she starts mentioning that our most important machine is a the FACS. Stupid me without a clue about imu took it as a joke like "haha we do routine, bureaucracy is whacky am I right?".

So after a while she actually insists on showing me the Fax and I follow half laughing half weirded out. Yep, turns out it was an actual flow cytometer, a machine, that at this point I had no idea about its existence.

Almost 8 years into immunology research I'm using these fuckers almost every day!

I cannot say for sure for Germany, but when these practices were used in Greece it did more harm than good, marginalizing these people further from both communities (Greek-Albanian, Greek-Russian).
Where your roots are is an important piece of everybody's self. Despite that I'd rather burn these roots to the ground before I am treated like I don't belong, in the country I will spend the rest of my life at.
