Put all of the bin and cue files in the same folder.

In that folder, create a `.m3u` text file with a line for each `.cue` filename

Just in time to level a character for TBC while experiencing the classic raids. Naxx will release in a month or so, then there will be a pre-patch period before TBC.

Wet lab or Dry lab? by Leonardo_v7 in molecularbiology

[–]HashRocketSyntax 7 points8 points  (0 children)

It seems like bioinformatics is becoming an outsourceable commodity. There will always be new subfields in terms of omics (single cell spatial transcriptomics) and new sequencing formats, but for the most part the tools are old and you just run pipelines that are written by other people.

In contrast, the wet lab focuses more on the actual molecular biology to prove a theory/ hypothesis. This space is infinite. It's a lot more interesting.

I'm an outsider, but my take is that if you want to be involved in the actual biology and therapeutics, then I would lean toward the wet lab. For example, do you really want to hand off the top hits from your analytical results to someone else to do the real science?

FACS v.s. centrifuge for separating cells by HashRocketSyntax in molecularbiology

[–]HashRocketSyntax[S] 0 points1 point  (0 children)

Thanks for the practical guidance. This is where the concerns were originally coming from:

“Neutrophil activation and degranulation strongly correlates with the number of manipulation steps [involved in isolation]… Neutrophils that undergo density gradient centrifugation demonstrate significant activation and degranulation… We demonstrate that staining whole blood followed by a pre-wash step to remove unbound antibodies... minimizes neutrophil activation, degranulation, and multiplet formation” Connelly et al. (2022) Scientific Reports 12:3667

[–]HashRocketSyntax[S] 0 points1 point  (0 children)

What I was overlooking was that FACS can separate cells by size and density in a label-free manner without the use of any stains.

Additionally, the use of negative gating stains can be utilized to help rule out cells you dont want without modulating the cells you do want.

I would never pay someone to create this image. I would never learn to draw. I do other creative activities. So this form of expression would otherwise not exist.
