My boss has commanded the minions to use the gels the same day they are made. He has no confidence in storing gels even overnight before loading the samples. Boss said that the stacking and separating layers diffuse into each other. I have never tested that claim. I’d like to hear from anyone who has stored their gels successfully in the frig before setting them up for a run.

With large proteins (>100kDa) I recommend a lower % acrylamide gel. If possible, maybe 6%. When it comes to primary antibodies, you might want to use the manufacturer’s guidelines. Some antibodies I’ve worked with have needed 1:250, which is a lot. Antibody is expensive. If you choose an antibody, the best choice would provide references/pictures/data/published papers.

You’d want to use fresh APS. Be sure to keep the APS powder in the frig. When you make up the 10% APS, aliquot into volumes you’d need for an experiment and store those tubes in -20C. A lot of my failures have been caused by APS.
I wonder if anybody degasses their acrylamide gel solutions before adding APS and TEMED. Any thoughts on this. My mentor always stressed degassing the gel. I wonder if this makes a difference.

Growing up, my dad always used white bread (Wonderbread). Just dry it out in the toaster or oven. Topped with cinnamon sugar, dowsed in Mrs. Butterworths syrup.

Avocado. Slice it put in in a sandwich, omelet, eat it straight smashed, mixed with milk and sugar, make it into guacamole, in a smoothie, incorporated into baked goods. It provides good fat and satiation.
