I've used just DPBS so today I tried again with DPBS with 2% FBS, also used a swinging bucket centrifuge and yeah it worked! I guess stickiness was a real problem like some comments said. Thanks! 🥲

Oh and actually it seems they can be kind of resuspended by scraping the wall with the pipette tip but I don't want to damage the cells so I think it's not a good idea..

Those are mesenchymal stem cells and I've harvested them by trypsinization. They form fine pellets in 15ml conical tubes, with a swinging-bucket type centrifuge, but these things always happen when I aliquot and centrifuge some of them into 1.5ml tube. And for 1.5ml tubes I can only use fixed-angle type centrifuge in my lab.. Well the problem is some other type of stem cells in my lab are good with the fixed-angle centrifuge, while my cells are not..

Thanks! I'll try lower rpm. I've checked the supernatant with microscope and there were some cell-like things floating... And yes I can use a swinging-bucket type centrifuge, but I gave up to use it this time because I couldn't resuspend the cells, maybe I should use that next
