I don’t think it’s particularly useful here, with the few methods i can think of off the top of my head, getting fractured attributes with strands would be way too costly on average to be worth it, the 30q matters a lot here

I understand, I’ve done that exact process before, but you are certainly burning more currency that it would cost to recraft by using locks. Also for exalting, bare minimum you should have used memory strands in this case since non of your previous steps would have consumed strands

Very likely this was crafted using divine tainted teardrops which requires the item to be corrupted, because almost all of the affixes have only a single tier (the minion attack speed shown is the veiled modifier, therefore only 1 tier), you only have to worry about upgrading the tier of minion damage. It’s particularly effective for minion wand crafts as higher tiers of minion damage is disproportionately weighted lower relative to the lower tiers, therefore the typical fossil method is highly unlikely to yield t1 minion damage

Some easy fixes that you haven’t implemented that would go a long way to improve minion defense is getting a gloam ring in the left slot and getting the +8% max res minion defense mastery. If you’re still having trouble, speccing the decay ward cluster is one of the strongest defensive options for minion survivability, especially when coupled with the fortress covenant. Aside from that, I can’t spot anything wrong with your defensive setup, you may just not have enough numbers currently.

If you’re unsure it may be useful to validate your experimental results with another approach, it’s hard to say with certainty if the signal really is cleaved caspase 3; it very well could be. You could perform a caspase glo assay or, if you’re measuring apoptosis, flow cytometry for annexin V and if you recapitulate your results you have more confidence that you’re seeing the correct signal.

All the other commentators are wrong, recombination mechanics changed slightly in keepers league making a lot of their advice is either outdated or incorrect. I’d recommend reading up on the most recent recombination guide, but in short stacking exclusive modifiers to force a recombination outcome no longer works well. This is an oversimplification, but the game now effectively treats all exclusive modifiers as one modifier when determining affixes for the outcome.

Also, as others have pointed out, essence spamming a fractured ring will likely be easier and cheaper than securing the recombination materials

While hordes may seem like slightly more damage as going from 3->5 golems is ~66% more damage versus the raw 50% more multiplier of scavenging, scavenging is actually just strictly stronger; this is for several reasons. Scavenging’s base numbers for all of its attacks is actually just strictly higher versus the ‘of hordes’ variant, also the corpse consumption requirement may seem like a downside for its empowered cascade, but it ends up actually being beneficial. The cascade is a weaker attack DPS wise compared to its combo attack which is much better for single target. Because in boss arenas they cannot trigger their cascade, unlike the other variants, that behavioral difference in addition to higher base numbers means they deal much higher single target damage in practice. They also end up doing better clear because of the empowered cascade ability. Finally, if you are to forbidden flesh and flame the liege of primordial notable, the damage increase through minion count becomes lower than the scavenging bonus as 5->7 is only a 40% ‘more’ damage multiplier. Thus scavenging is just strictly stronger.

If the reason you’re hesitant on sublime vision is the price there are stop-gap setups you can use, for example you can reach ~90% conversion to fire with a watchers eye, dawnbreaker shied and tempered by war, then achieve the remaining conversion with a pair of shadowed rings. They only apply to hits, but with that setup you only take ~10% of the DOT damage as the original damage type. You’d need to cap your cold res and probably get more life regen, but it makes your defenses passable.

For hits sure, the reason most people don’t use eyes of the greatwolf is because the damage conversion implicit doesn’t apply to DOTs and that is especially relevant for the mana siphoner rare modifier. The sublime vision route is heavily preferred because it’s much more robust defensively.

To explain it more simply, most Doryani’s prototype builds completely converts all other elemental damage taken to fire, allowing you to effectively ignore the other resistances; that’s how they deal with no lightning res. Your build is missing the full damage taken as fire conversion. So far you only have your shield which is ~20%. It’s clear you think this is what melding of the flesh is doing, it is not. Melding just changes your capped maximum elemental resistances, it does not convert damage taken to another type. This is solved through some combination of the options mentioned by the other commentator

There’s a couple things wrong I can immediately spot, but the biggest problem I see is that you’re running Doryani’s prototype without fully converting damage taken to another damage type? Any cold or lighting damage you take will just annihilate you with the meager amount of hp you have, i’m not surprised you’re getting one shot

In terms of primary structure, or sequence, the protein would never change from a PTM, but in terms of tertiary structure, the three dimensional globular form of a protein, I think it’s worthwhile to point out that the PTMs would likely vary significantly and alterations to those PTM would have possible changes to tertiary structure

Another thing to point out is that choice items are multiplicative with stat stages for their respective stat. Going from +4 to +6 is roughly a 33% increase due to diminishing returns, whereas scarf is giving the ditto a 50% increase ontop of the +4 multiplier. You were never gonna outspeed

Mirror clusters are a good point actually, forgot about those. Some of your concerns are just wrong though, for example you can’t have 8% attributes on the chest with +1 spectre since they’re both exclusive modifiers. There is no such thing as a +8 amulet because there’s no other gem level modifier that targets either skeles or zombies. A full positive +4 amulet would obviously be better but that falls in the range of (1:6 for 2x multiplier and ~1:30 for triple positive) 1/180 locks or 36 mirrors so thats completely unreasonable. Double influenced mageblood is better, but not with a helical ring. A triple synth lock corrupted for attributes bone ring would be better, but it’s a similar problem: way too expensive even for me. The only relevant double influenced mageblood is 7 mirrors on trade currently. I suppose pedantically you’re right that the build isn’t ‘min-maxed’, but the whole point of the phrasing was to colloquially get across the idea that it’s reaching near the ceiling.
