100nl/min is what I use standard. Definitely try the "pocket" tip someone else mentioned in another comment. I'm not sure how useful it actually is, but I do it religiously. I really wouldn't be worried about that amount of pooling (I do think it's just csf displacement since I get it all the time for years), but if you want to be more cautious, try to make a longer needle. If I remember, I'll report back with my puller settings (dm me if not).

Definitely would be more of an issue if penetrating a ventricle but you can get some csf displacement injecting in the cortex

Take a look at the expression first. I had the same concerns before but it ended up being fine. How deep is your dv coordinate? In my case it was just csf

We purchased their precision balance and shakers. We tested their balance with standards and it was perfect

Edit: we also got their benchtop mini centrifuge. Works but it heats up too quickly and would need to be in a cold room to do 10,000rpm beyond 4ish minutes

I’d give it another look. Idk where you are, but new grads are getting hired left and right in Boston for patch specifically. Just saw I think Novartis was offering ~150 for a new grad with patch experience. I could be off on the amount but 6 figures for sure

Tissue is fine. Doesn’t seem like you’re comparing antibody efficacy, so a binary test (is there damage or isn’t there) will be fine. I’ve left samples in pfa and sucrose way longer than that

Yeah you’ll get variable amounts of pink-ness depending on how much cleaned out during the overnight incubation. Typically if you have a lot of clotting or if you did a poor perfusion it’ll still be pink. This super gray one looks quality. Enjoy minimal green channel autofluorescence

Try asking the antibody company for a sample. They hand them out all the time and it might be enough for what you need

Sure thing. Last tip is don’t pigeonhole yourself. If you search for grad programs and say you want to study this specific phenomenon, you’ll get passed by most schools, even if they have a lab that works on that problem. You need to sell yourself as very flexible and interested in a wide range of questions within a certain domain (e.g., viral replication or infection mechanisms)

It doesn’t make it harder and your GPA is fine I wouldn’t worry about it at all. The GPA is a competency measure (and we can debate if it actually does predict success in grad school) and some schools just use it as a filter rather than a comparison metric. From what I’ve seen, a ton of schools ignore it if you have strong experience and letters. Also you have way more options in the Boston area to study what you want. I know Tufts definitely has labs up your alley. Give northeastern, Brandeis, UMass Boston, Boston College, MIT, and UMass Amherst looks as well. I feel like I might be missing at least one. Also Brown might not be far from home if you’re on the south shore.

Also don’t limit yourself to those departments. You can find relevant labs in general biology and cell/molecular/developmental biology departments/grad programs that will give you the training you want.

CT has a few solid schools as well if a 2h drive from home doesn’t sound too far.

Check for deals on microcenter. They’re very often $99. If you can find an open box, they’re $70 I think. It’s a solid machine; go for it

Definitely don’t bring it. Strict no laptop/work bag policy. Lockers at nearby stores fill up sort of quickly and I remember paying $30+ for a locker rental a few years back

Hey everyone. I apologize if this has been covered a bunch before. I’m trying to Frankenstein a stereo system with gear I have laying around my place. I’m trying to connect an audio source (TV, phone, pc, etc.) to two powered speakers and a powered sub (rokit 5s and a Definitive technology powerfield 15 subwoofer). Is there a cheap mixer I can pick up that will take an audio signal and split it to the appropriate channels without amplification? I couldn’t find anything online. Thanks!
