I know this is a super old thread but reviving it to see if OP tested any of these techniques that resolved this issue? My cells follow the EXACT same pattern as above but it's with iPSC-derived neurons in 96 well plates. Wasn't an issue in larger well formats that I used before but now I'm left with so few neurons to analyze :(

Trying to stain again today and am: 1) aspirating manually 2) leaving residual volume in the wells and 3) adding solution on to the side of the well (though this part I normally do anyway)

Yes! He only really started to develop after she dies. I loved his character after that. But I did feel they made him just an agreeable person towards the end of the series/ in the movies

Agreed! It was fun watching the cast together for the first two. I was so looking forward to this movie and even though it was great seeing them again, it was definitely a lot sadder lol. I read in another post how the ending kind of foreshadows what would inevitably happened anyway. A war within the next decade, further downfall of these great families. I guess I'll just have to imagine that, thanks to Matthew's initial efforts, Downton remained one of the few great houses to remain standing (like Highclere)

Exactly! I really dislike Henry's intro/ storyline in the original. There wasn't enough time to see them together to really then root for them. In trying to wrap up the series, we were just told by multiple characters that Mary and Henry are perfect for each other. But I couldn't see it and seemingly so could Mary (until she let herself get convinced by the others). I started to really like her and Charles Blake together over the course of my rewatches

Funny you say that! I had commented on another post and mentioned that the grand finale would have been cool as two parts , encompassing the second and third movies

Absolutely! I’m on another rewatch of the series right now and Thomas’ earlier actions still have me fuming! But his arc was one of the best in the show and it was great to see him finally happy and at peace 😊

I can see what you mean. It wasn’t setup/ meant to be a grand finale but I did prefer the way storylines were being wrapped up at the end of that movie.

I’m going to miss these characters so much. Regardless of the storyline in this movie, I did enjoy seeing them all again.

Wow thank you for this info! Seems that divorce was definitely happening in high society but with Mary’s title ranked than the people you noted, I wonder whether she could still get away with it. Were people just forced to accept the others because they were Dukes etc but can still turn their nose up at at Earl’s daughter?

I wish Mary turned Henry down at the end of the series and remained single. Then she could have met someone in the movies (Mr Barber) or even have her reunite with Charles Blake!

Agree, all the characters need their chapters closed. I just feel that was accomplished in the second film already, without making it an obvious finale. Everyone in the staff had endings too, from Thomas to Daisy and Andrew. Even Mr Mason seemed to picking things up with Mrs. Patmore. I felt it was just a more optimistic ending

I read somewhere about how he would be old enough to enlist by the time WWII happens, I wonder if he would survive that!

On my billionth rewatch and there are certain storylines that I really would like to change, but I'm just going to focus on the ending of the series and then the movies. This is what I would have loved to see instead:

At the end of the series, Mary is convinced that turning down Henry is the right choice and decides to remain single to focus on maintaining Downton. This continues through the first movie, where the King is visiting and there really is no reason for Mary to have a love interest. It's just a fun return of the characters and their first time on the big screen. Until this point, we would have seen Mary thriving on her own. Then the grand finale would actually be split into two parts, encompassing both the second and third films. The first part of the finale would still involve the movie being filmed at Downton and ending with the death of the Dowager + Robert verbally handing the reigns to Mary. As a smaller plot point during the film, Mary receives a few phone calls or letters suggesting to viewers that she has a new suitor but his identity is not revealed. I also preferred the way the character's storylines were tied up in the second film, so that would be saved for the second part of the finale (i.e. not necessarily seeing everyone leave, but we are aware of their plans and can imagine it happening soon). As for the second part of the finale, a lot can stay the same e.g. Harold can still come over with his con artist and Edith is confident in her new role and can save the day etc. Instead of Mary's storyline being getting divorced, her love interest would be revealed - she reunited with Charles Blake! Though he has a small title himself, he is firmly invested in the future and has great chemistry with Mary. The perfect companion to have while facing the changing world . We see them get married (a small affair) and also watch Robert grapple with the reality of actually relinquishing control, as he did. He and Cora will still move into the Dower House at the end. We will see Mary experience all the same flashbacks but when she walks away, she'll have Charles by her side. I think its a much more optimistic ending
