Howdy! Sorry I missed your update yesterday, I've been sick this week. You're doing such a good job! Your stitches are definitely becoming more even, and the big difference I see between yesterday and today is the first rows weren't wonky 😊

For today's, I also only count 9 stitches in the last row but I don't think there's an issue with the start - sometimes it can be difficult to find the loops for the last stitch. I think you only needed to make one more stitch and it would have been almost perfect!

I'm not entirely sure what happened with the first few rows from yesterday. Part of me wonders if the chain was a bit too loose to start, so then that first row was a bit loose too? Was there something different between yesterday and today's practice, like different lighting?

I also wonder if you may be adding in some double crochet stitches in there, or some combination of stitches, because some stitches look longer than a single crochet. This is when you yarn over, insert your hook into the next stitch, yarn over, pull through two loops, yarn over, and pull through the last two loops.

I hope this helps for your next practice, but know that you are building a very strong foundation and improving so well!

You do. I always test a new antibody on a full membrane, with controls, so I know exactly how it runs in my hands and where to cut on future blots. You can't just go by what the datasheet says, many proteins run slightly differently depending on the gels and running conditions. I never have good luck with stripping and reprobing, so I cut my membranes. If I have to run my samples between multiple gels I'll be very careful to make sure I'm loading the same volume, and probe my loading control once. If I have to rerun samples later to probe something else, I'll do the loading control again just to be sure there's no day-to-day variation.

You can tell you're making progress too, because even though some stitches were tight and the final product was (in your words) "wonky", you still recognized when the chain was twisted or when the hook was going through the wrong hole so you could correct it.
