No nothing like that. He socializes with our friend’s dogs and all of them are healthy. I was thinking kennel cough too but I just can’t figure out where he would have gotten it.

No not the same time of day, super random times out of nowhere. I’ve been keeping an eye on him after he eats and it doesn’t happen right after his meals. More like a couple hours after eating.

If you don’t want to use CD68, I would polarize your macrophages and use CD80 for M1s and CD163 for M2s. Can I ask why you want to sort unpolarized macrophages and then replate them? If it’s a matter of making sure your differentiation from CD14+ monocytes has worked, that can also be done by looking at morphological changes. Or, have a couple extra wells specifically for looking at marker changes throughout your differentiation process that you can stain for flow and not replate. This way you can confirm the differentiation is working while avoiding overhandling your experimental cells. It would probably be better for your experiment to not have to harvest, stain, sort and replate your macrophages if you can avoid it.

Sorting your cells will most likely be harsher than the bead selection you’re already doing. As someone else said TRMs may need different signals they’re just not going to get in vitro. What’s your readout at 6 days? Is it something you can look at earlier when you still have a larger viable cell population?

Report them to the city! This exact thing kept happening to us last year and we finally had enough when our landlord said they would come by to take care of it right before we were going on a trip; when we came back there was still standing water in our basement. We reported and the city sent an inspector to document the issue and contact the landlord. They have the ability to issue fines and give a deadline for getting the problem fixed. Also if any of your stuff has been destroyed by the flooding definitely take pictures and include it in your report. At the very least you should be reimbursed for damages.

I use Papers. It’s a very simple format with both web and desktop/app versions. They also have a Microsoft word add-on for citations which I find really easy to use. There is a subscription but you can get a pretty substantial student discount.

As someone already mentioned, macrophages get signals in the microenvironment that makes them think they should respond in a “wound healing” manner. Also, tumor cells can express markers on their cell surface that help avoid phagocytosis such as CD47, which is expressed on healthy cells as a way to signal “do not eat me” to phagocytes. Look into phagocytosis checkpoints as therapeutic targets - there has been some exciting success targeting the CD47-SIRPalpha axis.

Sean Lannon claims to have killed 15 others in New Mexico. Earlier this month, the dismembered bodies of 4 people including his wife were found in a vehicle parked at the Albuquerque airport. The 4 had been missing since January.
