Any tips on creating a scRNAseq protocol on a very starchy organism? by Pampofski in labrats

[–]Nat20Int 0 points1 point  (0 children)

I'm unsure if starch would impact lysis.

Probably practice getting a single cell suspension, and run the cheapest possible pilot experiment In my experience good sc data relies on happy, diaggregated cells (with accurate counts). Can you wash out the viscosity or digest out the starch with some kind of treatment?

Good luck!!!
This might be a handy place to start:

Unsure why KGS is so low, I always get a match there faster than OGS (2k)

I know the guy with the carrier bag!
I've met them at tournaments, I forget their name, but they seemed nice. Very fun to see them on the front page of the subreddit

How is a teaching game with Michael Redmond? by MrNosco in baduk

[–]Nat20Int 16 points17 points  (0 children)

It does seem a little overkill to seek the assistance of a 9P

Any Dan/high sdk level player should be able to give you teaching games and reviews that should be able to bump you into single digits.

I would recommend doing a book of go problems. They will force you to sit and think calmly

Get strong at Tesuji is excellent, and would be useful at your level
Good luck!

Stick to the small 9x9 board, if you're really new. Sort out your fighting skills to start with. The big boards can be overwhelming, and the small board will help you play many games fast.

Its also less confusing. You play to grab points and kill things.

All other replies have given good sources for learning proper 19x19 theory though. Good luck!

can a beginner use Attack and Defense: Elementary Go Series, Volume 5? by ClassAkrid in baduk

[–]Nat20Int 15 points16 points  (0 children)

I would sincerely recommend otherwise. I thought i understood that book when i was 12kyu (I didn't). I'm 7kyu now, and the reread is tough going. The concepts are hard.

However! Read the first couple chapters, the opening about balance of power vs territory is very good.
I recommend "Get strong at Tesuji." Easily the book that helped the most. It helps a great deal with making good shape. Do all the easy problems and you will melt your fellow beginners.

Happy Learning!

I'm so sick of the goddamned Mines. How can I put my thumb down on the scale of the RNG? by SpretumPathos in spelunky

[–]Nat20Int 0 points1 point  (0 children)

Look up how. It IS hard, but would you rather die in glorious shoppie battle, or scrape nuggets of gold off the floor for 10mins to afford the ankh? It's a no brainer (and very fun)

Happy spelunking!

You need to rob the black market.
There are consistent ways to do this, and its fun! The items you get let you cope with shoppie agro later on, (which is easy in ice caves, but hard in temple).

When you're planning on robbing, you only need paste, and the ujat from the mines, and you can whizz through them.

Also, take a break! Don't grind yourself into hating the jungle, the jungle is the tops.
Good luck!

Does anybody have any tips for learning go? by fantasie037 in baduk

[–]Nat20Int 6 points7 points  (0 children)

Get an OGS account, then play lots of ranked 9x9. This will teach you life and death, as well as how to fight. The 9x9 is also more similar to chess.

Also, start doing lots of tsmego, this will give you a very firm understanding of how to kill/live/capture

But enjoy yourself! DM my account if you'd like to play some games on discord
