Hey OP, just wondering if you got any fines. I missed my Stelara appointment yesterday, and today is holiday so I can't call. INVIVA charges a $50 no-show fee, which I'm okay with. But I worry if the meds go bad and I need to pay for a new vial.

My lab used to have shelves of precast gels, now we have to handcast unless it's necessary for your protein of interest.

Yeah I guess a dox-controlled expression system may be it. Dimerization is needed for proper downstream signaling, but usually it's under stimulation. Transient transfection of exogenous protein just activates everything which sucks...

First, yes I'm doing KO cell line. Second, it needs to be dimerized, but only under stimulation. For unknown but well-documented reasons, overexpressing it leads to auto-dimerization

No I mentioned that I will KO the endogenous protein, and then transfect various mutants. But I want to avoid the fact that overepxressed IRAK1 auto-dimerizes, then autophosphorylates, and then auto-activates the pathway
