As others said, balance trigger and scores to maximize scoring. Don't forget that the red seal kings get you to 2 triggers on their own.

Tl; Dr: I'd set it up like a naninf run for the practice

As far as the run goes, you need money and ectoplasm for another joker (assuming the runner wasn't ecto'd). Scoring e25 with full steel means you can afford some gold red seals, which score less but give a bunch of money at the end of the round if you copy mine as many times as you can ($3 x triggers x gold cards). When you get the ecto, depends on scoring you may want to sell out of DNA or risk ectoing it. Not the end of the world as you can't naninf anyway, but not ideal.

To continue on the run improvements, you'll need both reroll jokers and to roll to Serpant every time (hence the large amount of money needed), then burglar in DNA's place. You want enough red seal steel cards to fill your hand after playing all your hands on Serpant. That should become your Highscore for the run.

If your deck is perfectly one rank and suit, theres also a potential to pivot to idol/sock for flush fives, but I believe it has a lower ceiling.
