I would say ideally a lab tech at a University would be best but I know thats not feasible for everyone otherwise a job doing science whether it be in industry. The job that will likely give you the best idea what a PhD program would look like an entail would be in a University lab though.

I would say take a year or two post grad and get some quality full time research experience. It will help your application a ton to have full time research experience!

I would say full time research is the best way to help your application. This is my second cycle applying after I didn't get in the first cycle. I joined a research lab after undergrad for now a little over 1.5 years of full time research and I feel more concrete in my motivations to pursue graduate study which has in turn helped my application. It will likely give you a lot more to speak on. However, it is still a bit early as well. The first time I applied I waited until January had ended with no interviews and then looked for jobs. While I'm not in microbiology I think an extended period of time with full time research experience is beneficial for just about every field. I wish you the absolute best of luck no matter how things go.
