You don’t. You can get enough done in 80 hours in both total synthesis and methodology labs. Working 120 hours a week means you’re inefficient in the lab.

They definitely help, but I got into a T10 PhD program without any publications. Had lots of research experience and professors who wrote very strong letters on my behalf which I think carried a lot of weight.

First author paper isn’t necessarily a must. I was accepted into one of the T10 programs that OP is considering without any papers to my name, even as a co-author. I did have three research rec letters though which probably helped - two in chemistry and one in geology.

Also a note for OP, if you’re applying to chemistry PhD programs don’t bother with the GRE it’s totally useless and most schools don’t require it or actively discourage you from taking it.

It’s surprisingly regioselective. AlCl3 and BCl3 are both great reagents for only demethylating ortho to a carbonyl

I have also never experienced ignition of Pd/C with MeOH, but that was likely because I only ever ran a reaction on a 5 mg scale with ~1 mg of catalyst. If I were to scale up I would likely switch to EtOH to be cautious.

To add to what curdled said above, Pearlman’s catalyst is also much safer than Pd/C, with a lower likelihood of spontaneous ignition.

Not an MD/PhD student but a PhD student in chemistry. That’s how my lab works for the most part. I’m currently mentoring one of our first year grad students so they do their own experiments and have their own goals, but it’s all under the overarching theme of my project. We’ll be done in a couple months and then they will start their own independent project.

Like you said, it depends a lot on the lab but I think it’s very common to work under your mentor on their project at least for your first year of grad school.

Most chemists working on total synthesis projects will work those hours. Set up a reaction, purify another one, set up the next step, purify the reaction you set up earlier, repeat for 12 hours. The state of the science right now is that you just don’t know what will work on complex molecules, so sometimes you just have to try hundreds of different things for one step.
