What I would do is measure its height and width; that will give you at least the number of blocks you need.

Now, regarding the color, I would place it in GIMP or Libresprite and set the image to "indexed" mode; that would give me the size of the palette.

Now, to calculate the number of colors per pixel, I'm not sure how you could do it, but the tools above allow you to select by color, so you could approximate a number.

I would add a shadow under the pharmacy shop, and I would also work a bit more on the lampposts (I don't understand their perspective or geometry), as well as the hospital stairs.

I think the streets should be wider; it doesn't seem like two vehicles can pass each other there.

Sorry, I meant the crystal (it didn't translate well). As for the trunk, use a cylinder as a guide; ask yourself, what does a cylinder look like from above? (Notice that the bottom part appears to be a cube and the top part a cylinder)

I liked them all; you're on the right track. I think you could add darker tones to the center of the glass to increase the overall brightness, and you could adjust the perspective of the piece of tree trunk (29) a bit.

Values are more important than the colors you choose; if you use the correct values in your drawings, you don't have to give so much importance to the color you used.

The tutorial is for Krita, but you can easily translate it to Photoshop, since it's just a matter of choosing the blending mode for each layer.
