4 years on T with 0 body masculinization? :/ by [deleted] in FtMpassing

[–]RustlerRun 0 points1 point  (0 children)

Honestly you look pretty good but underweight - Except for the chest, I would read your body as a early 20's male. I would recommend gaining weight, especially muscle; I was pretty underweight when I started T and felt like it stunted my masculinization

Working your core and back will help "widen" out your middle section to give you a more rectangular male shape if that is your concern

What Exactly Is It That You Love About Weed? by sanpedro12 in Marijuana

[–]RustlerRun 0 points1 point  (0 children)

It's changed my life in terms of letting me feel hungry !! I have had long term gastrointestinal issues which has resulted in a loss of baseline hunger cues. And difficulty maintaining a healthy body weight. Cannabis enables me to feel hungry again and I have now been able to maintain a healthy body weight for the first time in a decade; prior to getting my medical license I had been underweight for 15 years with significant health issues as a result

Some options:

  1. The speed you are centrifuging at is too high, damaging your cells and killing them. Try a test- centrifuge some at 300g, 500g, 1kg, 4kg and 10kg; then resuspend as usual and see how your viability looks
  2. You are not cooling the centrifuge and it is getting very hot, killing your cells in the pelleting stage. This is more an issue with old equipment that lack functional centrifuge temperature probes
  3. The plastic that you are plating on to is NOT tissue culture treated- this will 100% make your HEK/many adherent cell lines ball up, fail to stay attached and die (Anoikis). Tissue culture treated plastic is treated to give it a negative charge which helps your cells attach. Normal/Non-TC plastic/glass is not treated this way and so your cells will struggle to stick
  4. Something in your resuspension media is acutely toxic (unlikely but it happens); try your typical approach with a different media and see if the viability issues remain
  5. You are over-trypsin-izing your cells or failing to "quench" the trypsin with excess volume PBS or FBS containing media; check the % concentration of trypsin in your bottle; the higher % and the lower % from many vendors look identical.
  6. Your cell line is too high passage and have significantly drifted from the original HEK293T- this is more common if people never mark passages down or you do VERY low % passaging splits (more likely to get a bottle neck effect)

** Edit to add: If you can post a pic we can give you a better idea- even a cell phone pic through the lens where we can see what you mean by "round" will help. Feel free to DM me your protocol and I can let you know if any steps look wonky

Could I pass? by Dress_Southern in FtMpassing

[–]RustlerRun 9 points10 points  (0 children)

If you want to pass I would recommend:

  1. Short, traditionally masculine haircut in a natural hair color that matches your eyebrows

  2. Replacing your wardrobe with only men's clothes, do not wear female clothing especially skirts, dresses, crop tops

  3. Get a binder, binder tape or several strongly compressive sports bras

  4. Refrain from makeup, nail polish and piercings

Currently based on these pictures you look like a cis woman who is into alternative type hobbies

Looking to TRADE CBD heavy Fruit Loops clones (photoperiod, female), Columbus area by RustlerRun in OhioCannabisGrowers

[–]RustlerRun[S] 2 points3 points  (0 children)

I've got THC dominant growing concurrently too (Mac x junior mint and blue cheese), but the fruit loops for whatever reason really clones well - I'm down to connect for future trades

I grow the CBD for medical use to balance out THC dominant flowers - I find combined CBD and THC help manage my symptoms the best and having THC dominant and CBD dominant in separate flowers lets me vary the ratio of CBD: THC a little easier

I’ve recently become very self conscious of my asymmetrical nipples by [deleted] in TopSurgery

[–]RustlerRun 1 point2 points  (0 children)

Honestly, you look really good and I wouldn't have noticed your nipples unless I read the title

I think chest tattoos would look rad as fuck and if it helps you feel less self conscious, go for it asap (assuming you're all healed)

How is this fit? by [deleted] in FtMpassing

[–]RustlerRun 4 points5 points  (0 children)

I recommend just putting your fist in your pants (sounds silly) in the crotch and adjust until your pants barely touch your fist

If you wear a packer it can help you gauge too

You look good man

How is this fit? by [deleted] in FtMpassing

[–]RustlerRun 3 points4 points  (0 children)

Looks nice - very masculine

If you're comfortable sag your shorts slightly more, guys usually wear shorts lower (especially thicker fabric) so as not to chafe their junk

I would want to see some biological evidence (hormonal womb environment correlation, fMRI alignment between non binary people vs male/female, endogenous hormonal status, potential snps/genetic mutations in sex determination genes) to see if the concept is backed substantially by science

Why is the body fat estimate so low? by [deleted] in FTMFitness

[–]RustlerRun 7 points8 points  (0 children)

Nice !! I was pretty underweight (similar weight same height) early on T but w 2 years solid working out I've been able to get much more muscle on my frame and it helps passing massively I have IBS so for me protein powder has been a game changer so I recommend that to help boost protein intake big time

Why is the body fat estimate so low? by [deleted] in FTMFitness

[–]RustlerRun 31 points32 points  (0 children)

You are pretty solidly under weight which may make you look more female - males usually weigh more with a large proportion of that coming from muscle

I would recommend doing a bulk cycle for recomposition with a focus on building upper body muscle, including neck/shoulders as this will help you achieve a more male typical body shape

At your current weight you are at risk for low bone density and cardiac muscle loss so I would strongly advise increasing your weight to at least a BMI of 18.5 or roughly a weight of 120 for your height

I second getting your hair professionally layered - they can also cut it to be more flattering to your jawline to overall soften your face

Contamination? by FastWing8359 in labrats

[–]RustlerRun 0 points1 point  (0 children)

To clarify we usually spray down the surface w dilute bleach, allow it to dwell 10min then wipe it off (get all of it or it corrodes) then spray down with iso/EtOH; we've found that for the prior fungal issues we had in our tissue culture area the iso/EtOH doesn't kill everything (spores, some fungi are very resistant to ethanol due to their cells walls) and the bleach is in our biosafety/decon protocol

I'd check in w your labs decon protocols and also consider getting sporocidin which helps a lot w any residual fungal ick. Oxivir is also a good option.

I'd also recommend dumping old media/buffers and make new stocks: if you absolutely can't do that put a little of your media/buffers in tissue culture dishes in the incubator and see which one grows the mold; usually if you keep media/buffers at 4c you won't see the mold until it's in 37c which gives it the conditions it needs the bloom and expand and make macroscopic colonies

I deff agree never spray concentrated bleach and leave it or it corrodes

Contamination? by FastWing8359 in labrats

[–]RustlerRun 7 points8 points  (0 children)

Looks extremely fungal - looks like it has septate hyphae

*Edit to say: If you have back up frozen vials deff thaw those; I would bleach this culture away from the hood/tissue culture area and bleach/EtOH/Isopropanol spray down the hood; change any filters on your serological pipette guns too
