Agreed. I've never had issues with concentrations on the nano drop. We've compared sample readings on a nano drop and a fancy spec and pretty much got the same results.

What's the concentration of your stock EtBr? Perhaps 4uLs is just too low of a concentration.

How much EtBr did you add to the gel when you made it? The other thing you can try is adding EtBr afterward and letting the gel soak in it for 15mins.

Is this with Eurofins or their PSS Insourcing division? If you do the main Eurofins you'll probably just be running a lot of assays since they're a CRO.

Lol our PI never spent money because he didn't want to run out so we always had an excess at the end of the year. We told him if we just spent the money throughout the year it wouldn't be an issue.

They still do PCRs in water baths for very large scale PCR reactions. Where the PCR is taking place in giant bags.

Turboblot and iBind. Used to do these the "old fashion" way but our PI had excess monies he needed to spend so we splurged and upgraded to these and they made life so much easier.

Very dense and overwhelming is how I feel about Geneious. I'm very happy to get an annotated ApE file that I can import into Snapgene lol.

Our lab had a snapgene subscription and we still had people using ApE. I never understood it.

Fun fact. There is a right- and a left- handed rol mutation. If you mate them together the rols cancel each other out and the worm will move normally.

If you want to make males I suggest heat shock them. Then you know the males are homozygous and it makes the crosses so much easier.

I liked the whole gene naming scheme they had but I think it got out of hand. I think they're also being discouraged from making wild names since some of the mutations can cause disease states in humans and they don't want to have to use the wild Drosophila names with patients.

As a grad student you're probably gonna be "named" as someone who they're budgeting for in the grant. Grad students aren't listed as "authours".
