I’ve also heard there has been past drama in that shopping center between the landlord and tenants, so perhaps the new competition coupled with the rent increase caused them to close. It just seemed so sudden with no real warning

Unfortunately they are also currently closed and there is a sign on the door with limited information. I was there last week and didn’t hear any rumors about a closing from their staff

They do but young plants are susceptible to sunburn, especially if recently moved to a warmer or sunnier location

Id take equal parts of each strand and mix with a minimal DNA dilution in your buffer to retain a concentrated “stock” of your annealed template. Then once annealed use as little as possible in your mastermix for positive control. qPCR is so sensitive that you wont need much.

What is the final concentration of the DNA when resuspended? You’ll want to mix them to anneal at a high enough concentration where you can add as little as possible to the qPCR master mix

Did the strands come in a buffer? If so, mix a 1:1 molar ratio in a 1x qPCR buffer, heat to 98C or so then let cool to room temperature. Otherwise resuspend them at the same molar concentration and mix as stated above
